Abstract

Therepeutic prepns. contg. insulin derivs., in which at least one of the NH2 gps. of the A1(glycine), B1 (phenylalanine) and B29 (lysine) amino acid elements is blocked by a substituent capable of forming an anion, and in which opt. at least one tyrosine OH gp. is blocked by a substit. easily removable in vivo, have a large proportion of the hypoglycaemic activity of normal insulin prepns. while being more sol. in H2O and having acceptable immunological properties. The OH-blocked cpds. extend the duration of hypoglycaemic activity. Pref. N-blocking gps. are succinyl and glutaryl, and OH-blocking gps. are pref. opt. N-substd. carbamoyl. The N-blocked derivs. may be prepd. by acylation with a suitable dicarboxylic acid or functional deriv. [FR2123524A1]
