Abstract

1280201 Quinoline derivatives F HOFFMANN-LA ROCHE & CO AG 2 July 1969 [2 July 1968] 33312/69 Heading C2C The invention comprises processes for the preparation of racemic or optically active quinoline derivatives of the Formula A wherein R 1 is H, OH, halogen, C 1-7 alkyl, C 1-7 alkoxy, or methylenedioxy, R 2 is vinyl or C 1-7 alkyl and m is 1 or 2, with the proviso that when R 1 is methylenedioxy, m is 1, and of acid addition salts thereof and the novel quinoline derivatives of the Formulµ Ib and IIb provided that when (R 1 )m is OCH 3 in position 6<SP>1</SP>, R 2 is other than ethyl, their antipodes and racemates and acid addition salts thereof. The said processes consisting of hydroxylating the corresponding racemic or optically active 4-(5- R 2 - 4 - quinuclidin - 2 - yl)methylquinolines or by cyclizing racemic or optically active compounds of the Formula E and if desired, converting the so-obtained bases into acid addition salts. Diastereometric 6 - methoxy - 4 - {3 - [ - 3(R)- vinyl - 4(S) - piperidyl] - 1,2 - epoxypropyl}. quinolines are obtained by deacylating diastereomeric 6 - methoxy - 4 - {3 - [1 - benzoyl - 3- (R) - vinyl - 4(S) - piperidyl] - 1,2 - epoxypropyl}. quinolines resulting from cyclization of epimeric 6 - methoxy - 4 - { - 3 - [1 - benzoyl - 3(R) - vinyl- 4(S) - piperidyl] - 1 - bromo - 2 - oxopropyl}. quinolines, which are made by brominating 6- methoxy - 4 - {3 - [1 - benzoyl - 3(R) -vinyl - 4(S) -piperidyl]-2-oxopropyl}-quinoline. Pharmaceutical compositions, suitable for oral, parenteral or rectal administration, contain the above novel quinoline derivatives of Formulµ Ib and IIb or antipodes or racemates or acid addition salts thereof in association with compatible pharmaceutical carriers. The compounds possess antimalarial and antiarrhythmic properties. [GB1280201A]
