A process for the enrichment of a substance capable of forming a complex with a polypeptide which comprises linking a polypeptide by a covalent linkage to a carrier to form an addition product, introducing basic groups into the addition product, contacting the thus modified addition product in water with a solution of the substance to be enriched whereby a complex between the said substance and the addition product is formed, purifying the complex by washing, causing the complex to dissociate, and isolating the said substance. The substance to be purified may be an enzyme e.g. trypsin, chymotrypsin, kallicrein, plasmin, pepsin, rennin, ribonuclease, thrombin, amylase, papain, hyaluronidase, carboxypeptidase A or B, pancreatopeptidase E, penicillinase or cholinesterase, or an inhibitor such as kallicreintrypsin inhibitor ("Kuritz inhibitor") or the trypsin inhibitors obtained from pig and bovine pancreas, from seminal vesicles, from plant seeds or from potatoes. When the substance is an enzyme the polypeptide used in the process may be the corresponding inhibitor and vice versa. The carrier may be a resin containing anhydride or acid chloride groups or activated fluorine atoms, or carboxyl groups plus activated groups. The necessary basic groups may be introduced by treatment with N,N - dimethylethylene - diamine, agmatine, N - methyl- N<SP>1</SP> - (3 - amino - propyl) - piperazine, 4- amino - pyridine, 2 - amino - pyridine, or 3- amino 1 - cyclohexyl - aminopropane. Purification of the complex may be by washing with water and buffer solutions, and dissociation may be by altering pH or ion concentration, by displacement, or by adding urea or guanidine salts.
