Abstract

Radio-immunological assaying of thromboxane B2 (=TxB2) content of blood plasma is based on the determn. of antigen-antibody bondings, due to changes in bonding measured, using a tracer cpd. contg. a radio-isotope (i.e. a radio ligand) and a specific antibody, in the presence of a sequence of different dilutions of a known TxB2 standard and the samples to be determined. A calibration graph is drawn based on the standard dilution sequence and related changes in bonding and is used during the determn. of the unknown samples by reading off the concentrations as a function of observed bonding changes. - The essence of the method is that high specific activity thromboxane - B2-tyronine-methyl-ester deriv. which contains iodine 125 isotope (1) is used as a radio-liquid. This liquid reacts with a specific antibody in a buffer soln. contg. 8-aniline-naphthalene -sulphonic acid and in the presence of the standard dilution sequence contg. known concns. of TxB2, and of TxB2 free blood plasma samples otherwise identical with the unknown samples, and finally in the presence of the blood plasma samples to be determined.
