Abstract

Method for solubilisation and naturation of somatotropin protein (I) from refractile bodies (RB) of a host cell comprises contacting the RB's with aq. urea or dimethylsulphone soln. at a pH effective for solubilisation. The soln. contg. (I) is then contacted with a mild oxidising agent to form intramolecular disulphide bonds between cysteine residues. - Pref. aq. urea is used, pref. at 1-10M, at above freezing pt. to below 25 deg.C and pH pref. 9-12. Pref. Tris buffer is used for pH control. If (I) is aggregated (oxidised in the refractile bodies, a reducing agent is pref. present, e.g. 2-mercaptoethanol.. Naturation is effected in the urea or dimethylsulphone soln. obtd., partic. in 3-5M aq. urea at pH 9-12 and at above freezing pt. to below 25 deg.C. Oxidn. is e.g. with H2O2 or air.
