Abstract

Prodn. of nourseothricin (I) or its salts or adsorbates is effected by culturing a (I)-producing Streptomyces strain under submerged aerobic conditions in a medium contg. C and N sources and mineral salts, with the following improvements. - (a) respiratory inhibitors of the alkali metal azide, pyrocatechol and/or ethyl isoamyl barbiturate type and/or amino acid transport inhibitors of the water-soluble Zn salt and/or beta-alanine type are added to the main culture; (b) the prodn. medium is heat-sterilised in the presence of phosphate-precipitating substances (esp. Zn, Fe, Al, Ca, Mn or Mg salts), sulphate ions and CaCO3, opt. with addn. of alkali, to give a pH of 7.2-8.2 after sterilisation. - (c) fermentation is effected with a phosphate supply of 0.1-10 mg/l.hr, a pH of 5-6.5, an O2 partial pressure of 30-80 (esp. 40-60)%, and with further substrate limitations, esp. with a wt. ratio of glucose units (as glucose or oligo- or polysaccharides) to NH4 N of 5-15; 0.015-0.2; (d) (I) is isolated as an adsorbate by adding an adsorbent, esp. bentonite, or is isolated by adsorption on a weakly acidic cation exchanger, elution with a dil. polybasic acid, concn., neutralisation, purificn. and pptn. in MeOH or drying.
