Ankrod (trade name: Arwin®, Arvin®) is an enzyme derived from the venom of the Malay rattlesnake (Agkistrodon rhodostoma). This enzyme is a highly glycosylated, medium serine protease with an average molecular weight of about 38,000, which has anticoagulant properties and is capable of dissolving blood clots.
Normal blood coagulation occurs by the action of thrombin, whereby thrombin cleaves fibrinopeptides A and B from the fibrinogen molecule, leading to the formation of fibrin (EP-B-0 556 906), which is a major component of thrombi in addition to red blood cells and platelets. In contrast to thrombin, the anode cleaves only the arginine lysine bond in the - ("A") chain of the fibrinogen molecule and thus releases the fibrinopeptides A, AP and AY [Cole et al., J. Vascular. Surgery 17: 288-292 (1993)]. The p- (B) chain of the fibrinogen molecule is not attacked and thus released by the anchor. Fragments formed after cleavage of the fibrinopeptides by the anode (des-"A" -fibrin monomers) can then polymerize into thin filaments. The atypical soluble fibrin produced is lysed by plasmin in the body and / or removed by the reticuloendothelial system (RÉS, a monocyte-macrophage system). The des "A" fibrinogen molecule is no longer cleaved by thrombin into natural fibrin because the resulting molecule is not a substrate for thrombin.
Anchor decreases fibrinogen concentration in the blood in a dose-dependent manner. Therapeutically induced and controlled hypofibrinogenemia reduces the viscosity of the plasma and the tendency of red blood cells to aggregate to such an extent that the blood flow properties are markedly improved, providing a prerequisite for increased blood flow in narrowed (stenotic) blood vessels.
Jelenleg például a krónikus perifériás artériás vérkeringés! zavarokat ankroddal kezelik, továbbá II fázisú klinikai vizsgálatokat kezdtek el ankroddal agyvérzéssel (gutaütéssel) kapcsolatban.Currently, for example, chronic peripheral arterial circulation! disorders are treated with ankrodis, and Phase II clinical trials have been initiated with ankroddal for stroke (stroke).
The anchor is preferably administered subcutaneously. Treatment can be performed in a hospital setting or, provided that regular monitoring of the fibrinogen concentration needed to monitor therapy is in place, or in an outpatient setting. Intravenous administration of the ankrod is also possible, but only in exceptional cases and under hospital control.
Az ankrod alapjában véve individuálisan adagolandó. Mértékadó a fibrinogénkoncentráció alakulása az ankrod alkalmazásától függően. A koncentráció lassan 70-100 mg/100 ml plazma értékre csökkentendő (terápiás tartomány). A teljes kezelési idő alatt a fibrinogénkoncentrációt ezen a tartományon belüli értékekre kell beállítani. A vér áramlási tulajdonságai ilyen feltételek mellett megfelelően jók. Basically, your anchor should be individually administered. Changes in fibrinogen concentration, depending on the use of the anchor, are of decisive importance. The concentration should be slowly reduced to 70-100 mg / 100 ml plasma (therapeutic range). During the entire treatment period, the fibrinogen concentration should be adjusted within this range. Blood flow properties under these conditions are good. Therapy usually lasts for 3 to 4 weeks, but may be extended if necessary.
Amennyiben a fibrinogénkoncentráció a terápiás tartományba esik, hetenként 2-3-szor 210-280 NE-t fecskendezünk be.For subcutaneous administration, 70 IU (International Units; 1 ml) is administered daily for the first 4 days, and 70-140 IU depending on trends in fibrinogen concentration from day 5 onwards. If the fibrinogen concentration falls within the therapeutic range, 210-280 IU are injected 2-3 times a week.
Intravénásán először 2-3 NE-t infundálunk testtömeg-kg-onként 8 órán belül. Ezután, az elért fibrinogénkoncentrációtól függően, az ankrodot utánadagoljuk. Általában elégséges minden 12 órában 1 NE/kgtesttömeg adagot lassan bevinni. For the first time, 2-3 IU / kg body weight should be infused over 8 hours. Subsequently, depending on the fibrinogen concentration achieved, the anode is dosed. It is usually sufficient to slowly inject 1 IU / kg body weight every 12 hours.
Az ankrod kezdeti felezési ideje a keringésben körülbelül 3-5 óra, ez azonban a koncentráció csökkenésével lassul úgy, hogy körülbelül 4 nap múlva, mely időn belül általában a beadott ankrod 90%-a eliminálódik, a felezési idő 9-12 napra hosszabbodik meg.The initial half-life of the anchor in the circulation is about 3 to 5 hours, but it slows down with a decrease in concentration such that after about 4 days, within which 90% of the administered anchor is generally eliminated, the half-life is extended to 9 to 12 days.
However, unlike heparin and warfarin, for example, there are fewer problems with non-specific bleeding during treatment with ankrod. See, Z. S. Latello, Thromb. Haemostasis 50: 604-609 (1983)], however, targeted treatment of such hemorrhages is still necessary and desirable.
Az ankroddal való kezelés kontraindikációi például a következők: haemorrhagiás diatézis (vérzésre való hajlam), valamint sérülések esetén, operációk után, szülésnél fennálló vérzés veszélye, fekélyes bélmegbetegedések, neoplazmák, rosszul beállítható magas vérnyomás, akut agyi Infarktus és tüdőtuberkulózis esetén, a RES-funkció zavarainál és az alvadéklebontás zavarainál, például magas lázzal járó állapotoknál, továbbá májmegbetegedéseknél, látható tünetekkel járó és fenyegető sokkos állapotok vagy terhesség esetén.Contraindications to treatment with an anode include, but are not limited to: haemorrhagic diathesis (predisposition to bleeding) and risk of postpartum haemorrhage after surgery, ulcerative bowel disease, neoplasms, poorly controlled hypertension, acute cerebral infarction and pulmonary tuberculosis, disorders such as high fever, liver disease, visible symptoms and imminent shock or pregnancy.
As described above, the risk of bleeding with an anchor is relatively low if the fibrinogen concentration decreases slowly and is adjusted to 70-100 mg / 100 ml during therapy. Patients with a history of latent bleeding, such as kidney stones or renal failure, should be treated with particular care. Arterial puncture and other drugs by intramuscular injection should be avoided. Caution should be exercised with concomitant administration of RES-blocking agents with ulcer drugs, anticoagulants, antifibrinolytics, thrombolytics and medicinal products that inhibit platelet aggregation and with intramuscular administration of the anchor. Absorption from muscle depression usually occurs very quickly, so that too much Des-A-fibrin monomer falls into the body and is at risk of thromboembolic complications.
Egy vizsgálatban, amelyet 429 betegen végeztek [Crit. Rév. Oncol. Hematol., 15, 23-33 (1993)], akiknél az ankrodot előzetes trombolitikus terápia nélkül alkalmazták, a vérzések összes előfordulása 9,8%-ot tett ki (4,2% belső vérzés; 5,6% külső vérzés).In a study conducted in 429 patients [Crit. Port. Oncol. Hematol., 15, 23-33 (1993)], in which an anchor was used without prior thrombolytic therapy, the total incidence of bleeding was 9.8% (4.2% internal bleeding; 5.6% external bleeding).

An antidote is currently used to neutralize the enzyme activity of the ankrod, which is based on a goat serum immunoglobulin preparation (Knoll AG, June 1983, title: Arwin®). This polyclonal antibody antidote is used for severe bleeding complications, or where there is an increased risk of bleeding, such as accidental injury, or when surgery is suddenly indicated. After the anchovy is neutralized, 4-5 g of human fibrinogen should be injected. If human fibrinogen, plasma or blood is used without prior neutralization of the ankrod with an antidote, there is a risk of acute disseminated coagulation.
Stocker et al., Thrombosis Research, 6, 189-194 (1975), examined thrombus formation in the presence of Arwin® alone and in the presence of a polyclonal antidote, and thus demonstrated the effect of the antidote.
In addition to polyclonal antibodies from goats, it is mentioned in EP-B-0 395 375 and EP-B-0 556 906 and in Burkhardt et al. (FEBS, 297, 297-301 (1992)) that monoclonal or polyclonal antibodies have been used to detect expression of anchor genes. Detection of fibrinogen in the blood using antibodies against ankrod and ankrod, or purification of ankrod using antibodies, has been reported.
A kecskeeredetű poliklonális antitestek ankrodantidotumként való alkalmazásának például az a hátránya, hogy ezek antitestek keverékéből állnak, amelyek közül soknak nincs ankrodneutralizáló hatása. A különböző antitestek sokasága gyors immunreakcióhoz vezethet, ezenkívül pedig viszonylag alacsony ankrodneutralizáló hatást eredményez. Ennélfogva az antidotum az ankrodhoz különböző affinitással rendelkező antitesteket tartalmaz. A poliklonális antitestek, mivel állatból származnak, csak rosszul standardizálhatok, azaz a különböző termelési sarzsok között ingadozások fordulnak elő.For example, the use of goat polyclonal antibodies as an ankrodantidum has the disadvantage that they consist of a mixture of antibodies, many of which have no anodrodneutralizing effect. The multitude of different antibodies can lead to a rapid immune response and, in addition, result in a relatively low anodneutralizing effect. Therefore, the antibody contains antibodies with different affinities to the anchor. Polyclonal antibodies, as they are derived from animals, can only be poorly standardized, i.e., fluctuations between different production batches occur.
The object of the present invention was to develop an antidote against the anchor which does not show the aforementioned disadvantages and is technically simple to produce.
Neutralizációs hatásuk a kecskeeredetű poliklonális antitestekhez viszonyítva in vivő legalább 100%-kal jobb. The object is achieved by a monoclonal antibody, antibody fragments, mixtures thereof or derivatives of the invention which bind to ankrodhoz, inhibit the activity and binding affinity of 1x10 -1x10 ~ _7 12 M range. They have a neutralizing effect that is at least 100% better in vivo than goat polyclonal antibodies.
Ennélfogva a kezeléseknél kis mennyiségekben alkalmazhatók. Advantageously, the antibodies used as the ankrodantidum of the invention exhibit a number of improved properties. For example: they form a homogeneous, well characterized product consisting of an antibody or a group of antibodies that do not show differences within the various production bands. These antibodies can be produced in any amount, and since they are not produced in an animal, they do not carry the risk of viral or bacterial infection. The antibodies, antibody fragments, mixtures or derivatives of the invention are epitope specific and exhibit high binding and neutralizing activity. Therefore, they can be used in small amounts for treatments. Due to the homogeneity of the product and the high binding and neutralization activity, the small amounts administered together have the importance of significantly reducing the risk of an immune response in patients. In contrast to those obtained with polyclonal antibodies, variations in binding and neutralization activity do not occur within the various antibodies, antibody fragments or derivatives thereof. A mixture of different antibodies, antibody fragments, or derivatives, whose binding activity is directed against different epitopes of the anchor, is very effective in neutralizing the anchor.
A találmány szerinti antitesteknek, antitestfragmentumoknak, ezek keverékeinek vagy származékainak az ankroddal szembeni kötési affinitása előnyösen 1 x 10-7_1 xio-12 M tartományban van, előnyösebben 1 xi 0“8-1 χ10-11 M közé és különösen előnyösen 1 κΐ o-9-1 χ10-10 M közé esik.Antibodies, Antibody fragments, mixtures thereof or derivatives thereof of the invention, the binding affinity is preferably from 1 x ankroddal 10-7_1 Xio -12 M, more preferably from 1 xi 0 "8 M -1 χ10 between -11 and particularly preferably 1 κΐ p -9 -1 χ 10 -10 M.
A találmány szerinti antidotumnak, ellentétben a kecskeeredetű poliklonális antitestekkel, in vivő legalább 100%-kal jobb ankrodneutralizáló hatása van, előnyösen 250%-kal és különösen előnyösen 500%-kal jobb hatása van. A monoklonális antitestek in vitro is jelentősen jobb hatást mutatnak, mint a poliklonális antitestek. In contrast to goat polyclonal antibodies, the antidote of the present invention has at least 100% better anodrodneutralizing effect in vivo, preferably 250% and particularly preferably 500%. Monoclonal antibodies also show significantly better activity in vitro than polyclonal antibodies.
Génmanipulált, rövidítetlen fragmentumok is előnyösen alkalmazhatók.The monoclonal antibodies or fragments thereof of the invention are in principle intended to include all classes of immunoglobulins, i.e., IgM, IgG, IgD, IgE, IgA or their subclasses such as IgG subclasses or mixtures thereof. IgG and its subclasses such as IgG1, IgG2, IgG2a, IgG2b, IgG3 or IgG4 are preferred. IgG / κ or IgG2b / K IgG subtypes are particularly preferred. A fragment is defined as any truncated or modified antibody fragment having one or two antigen complementary binding sites which exhibit high binding and neutralizing activity to the anchor, and antibody portions containing an antibody corresponding to a light and heavy chain binding site, Fv, - either F (ab ') 2 fragments or single-stranded fragments. Preference is given to truncated double-stranded fragments such as Fv, Fab or F (ab ') 2 . These fragments may be produced, for example, by enzymatic treatment, cleavage of the Fc portion of the antibody by enzymes such as papain or pepsin, and by chemical oxidation or genetic engineering of the antibody genes. Gene-manipulated, non-truncated fragments are also advantageously used.
The antibodies or fragments may be used alone or in a mixture.
Géntechnológiai manipulációkhoz az antitestgének a szakemberek számára ismert módon, például hibridómasejtekből izolálhatok. Úgy járunk el, hogy az antitesttermelő sejteket tenyésztjük, majd az mRNS-t ismert módon izoláljuk, azaz a megfelelő optikai sűrűsé3For genetic engineering manipulations, antibody genes may be isolated from those known to those skilled in the art, such as hybridoma cells. The procedure involves culturing the antibody-producing cells and isolating the mRNA in a known manner, e.g.
The cells were lysed from guanidium thiocyanate, extracted with phenol, chloroform / isoamyl alcohol, precipitated with isopropyl alcohol and washed with ethanol. The cDNA is then synthesized from the mRNA by reverse transcriptase. The synthesized cDNA is incorporated directly or after genetic manipulation, such as site-directed mutagenesis, insertions, inversions, deletions, or base exchanges into appropriate animal or fungal or bacterial or viral vectors and expressed in appropriate host cells. . Bacterial or yeast vectors pBR322, pUC18, pACYC184, lambda or yeast are preferred for cloning genes and expressing in bacteria such as E. coli or yeast such as Saccharomyces cerevisiae.
Előnyösek a hibridóma-sejtvonalak. Ilyen hibridóma-sejtvonalakat ismert módon, például a következőképpen állítunk elő: ankroddal immunizált állatokból antitesttermelő B-sejteket izolálunk, a sejteket ankrodkötő antitestekre nézve szelektáljuk, majd ezeket a sejteket, például emberi vagy állati eredetű, például egér-mielómasejtekkel, humán limfoblasztoidsejtekkel vagy heterohibridómasejtekkel való fuzionálás [Koehler et al., Natúré, 256, 496 (1975)] révén vagy e sejtek megfelelő vírusokkal való megfertőzése révén örökéletűvé tesszük. Előnyösek a fúzióval előállított hibridóma-sejtvonalak, különösen előnyösek az egér-hibridómasejtvonalak, egészen különlegesen előnyösek azok a hibridóma-sejtvonalak, amelyek a MÁK 1-2, MÁK 2-29/3 vagy MÁK 3-27 antitesteket szekretálják, és amelyek a DSMZ intézményben (Deutsche Sammlung für Mikroorganismen und Zellkulturen in Braunschweig) DSM ACC2317, DSM ACC2318 és DSM ACC 2319 számon vannak letétbe helyezve (MAK=monoklonale Antikörper=monoklonális antitestek). The invention further provides cell lines that synthesize antibodies of the invention. The synthesis, as mentioned above, can be accomplished after transformation of animal or fungal or bacterial or yeast cells. It is preferable to use hybridoma cells or trioma cells. Hybridoma cell lines are preferred. Such hybridoma cell lines are prepared in a known manner, for example, by producing antibody-producing B cells from animals immunized with an anodode, selecting the cells for anchor binding antibodies, and then, such as human or animal cells such as mouse myeloma cells, human lymphoblastoid cells or fusion (Koehler et al., Naturre, 256, 496 (1975)) or by infecting these cells with appropriate viruses. Hybridoma cell lines produced by fusion are preferred, mouse hybridoma cell lines are particularly preferred, and hybridoma cell lines secreting MAK 1-2, MÁK 2-29 / 3 or MÁK 3-27 antibodies are particularly preferred, and which are secreted by DSMZ. (DSM ACC2317, DSM ACC2318 and DSM ACC 2319 (MAK = Monoclonal Antibody = Monoclonal Antibodies)).
A fent említett hibridóma-sejtvonalak a különösen előnyös IgG típusú antitesteket szekretálják. The above hybridoma cell lines secrete particularly preferred IgG antibodies. The resulting MAK 1-2, MAK 2-29 / 3 and MAK 3-27 cell lines belong to the following IgG subtypes: IgG / κ, IgG2b / K and IgG / κ. These preferred antibodies bind to different epitopes on the anchor molecule, as shown by competitive binding studies between the antibodies. Binding of the particularly favorable MAK 1-2 to the epitope of the antibody results in the most efficient neutralization of the anode molecule, thus requiring the lowest amount of antibody to neutralize the enzyme effect. Monoclonal antibodies show a significantly greater neutralizing effect than the polyclonal antibody-based antidote commonly used in the treatment of bleeding, which is obtained from Knoll AG (Ludwigshafen) as an antidote.
Derivatives of monoclonal antibodies of the invention include peptides and peptide mimetics derived from the antigen-binding region of the antibody, as well as solid or liquid carriers such as polyethylene glycol, glass, synthetic polymers such as polyacrylamide, polystyrene, polypropylene, polyethylene, such as cellulose, antibodies, fragments or peptides bound to Sepharose or Agarose, or conjugates with enzymes, toxins, or to radioactive or non-radioactive markers, for example 3 H 123 |, 125 | 1311, 32p 35 S> 14 C> 51 Cr> 36 C | f 57 Co _ δδρθ, 59p e , 90γ mTc, 75 Se -, for fluorescent / chemiluminescent markers such as rhodamine, fluorescein isothiocyanate, phycoerythrin, phycocyanin, fluorescein, metal chelates, avidin, streptavidin or biotin, covalent antibodies.
The antibodies, antibody fragments, mixtures and derivatives thereof of the invention can be used directly or after drying, for example, after lyophilization, after binding to the aforementioned carriers, or after formulation with other pharmaceutically active substances and excipients. Examples of active ingredients and excipients are additional antibodies, antimicrobial agents which have a microbiocidal or microbiostatic action, such as antibiotics in general. Other examples include sulfonamides, antineoplastic agents, water, buffers, saline solutions, alcohols, fats, waxes, inert carriers, or other substances customary in parenteral formulations, such as amino acids or sugars. These pharmaceutical compositions are used in the treatment of diseases, in particular in the treatment of coagulation disorders, in particular in the treatment of peripheral circulatory disorders or in the case of chills.
The antidote of the invention may be administered orally or parenterally, either subcutaneously, intramuscularly, intravenously or intraperitoneally, with intramuscular or intravenous administration being preferred.

Ezeket a sejteket 5:1 arányban kevertük össze, 1,5 ml PEG-oldattal (=polietilénglikol-oldat) 1 percig 37 °C-on inkubáltuk, majd PES-t adtunk (1 ml-t 30 másodperc alatt, 3 ml-t 30 másodperc alatt és 16 ml-t 60 másodperc alatt) hozzá. Egy mosási lépés után a sejteket szelektáló táptalajban [10% FCS (fetális borjúszérum), 10% Condimed H1 (Boehringer Mannheim), HAT-kiegészítő (hypoxantin-, aminopterin-, timidinkiegészítés), ITS-kiegészítő (inzulin-, transzferrin-, szelenitkiegészítés), piruvát, glutamin, sztreptomicin/penicillin tartalmú DMEM (Dulbecco által módosított Eagle-táptalaj)] 37 °C-on, 7,5% CO2-ot tartalmazó atmoszférában tenyésztettük.Three days after the last antigen injection, the spleen was harvested, the cells were washed, separated, and the lymphocytes fused with the myeloma cell line SP2 / 0-Ag14 (ATCC CRL1581). These cells were mixed at a ratio of 5: 1, incubated with 1.5 ml of PEG solution (= polyethylene glycol solution) for 1 min at 37 ° C, and then PES (1 ml for 30 seconds, 3 ml) was added. 30 seconds and 16 ml in 60 seconds). After a washing step, the cells in the selection medium [10% FCS (fetal calf serum), 10% Condimed H1 (Boehringer Mannheim), HAT supplement (hypoxanthine, aminopterin, thymidine supplement), ITS supplement (insulin, transferrin, selenite supplement) ), pyruvate, glutamine, streptomycin / penicillin-containing DMEM (Dulbecco's Modified Eagle's Medium)] were grown at 37 ° C in 7.5% CO 2 .
A specifikus antiankrodantitestet szekretáló hibridómák azonosítását specifikus ELISA-val, mikrotitrálólemezen végeztük a következőképpen:The identification of hybridomas secreting specific anti-anchodant antibody was performed by specific ELISA on a microtiter plate as follows:
- Mikrotitrálólemezeket tartályonként 0,1 ml ankroddal, illetve a specificitás meghatározásához referenciaproteinekkel fedünk (1 pg/ml 0,05 M nátrium-hidrogén-karbonát, pH 9,2) 16 óra alatt, 4 °C-on.- Microtiter plates were coated with 0.1 ml of anchor per well and with reference proteins (1 pg / ml 0.05 M sodium bicarbonate, pH 9.2) for 16 hours at 4 ° C.

A pozitív antitestreakciót adó hibridómákat szubklónozással választottuk szét, és az egyedi kiónokat újból teszteltük. A legmagasabb reaktivitású antitesteket használtuk in vitro neutralizációs vizsgálatban. Ilyen módon számos pozitív hibridóma, azaz ankrod elleni antitestet termelő sejt izolálható.Positive antibody hybridomas were separated by subcloning and individual clones were retested. The antibodies with the highest reactivity were used in the in vitro neutralization assay. In this way, a number of positive hybridoma cells, i.e., cells producing antibody to ankrod, can be isolated.

Monoklonális antitestek termelése és vizsgálataProduction and assay of monoclonal antibodies
A monoklonális antitesteket szérummentes sejttenyészet-felülúszókból tisztítottuk. Monoclonal antibodies were purified from serum-free cell culture supernatants. To this end, hybridomas are stepwise from DMEM / HAT / 10% FCS to DMEM / HT / 10% FCS and DMEM / 10% FCS to serum-free cell culture media such as SF-3 (Cytogen), PFHM-II (Gibco), HL-1 (Bio-Whittaker), Ultra-Doma PF (Bio-Whittaker) or the like (HT = hypoxanthine, aminopterin). Subsequently, Protein A and Protein G-Sepharose were used for affinity purification.
Miután a sejttenyészet-felülúszókat felvittük a kromatografálóoszlopokra, az aspecifikusan megkötött proteineket 3 M nátrium-klorid/1,5 M glicin (pH 8,9) oldattal mostuk ki; az antiankrodantitest-aktivitást 500 mM nátrium-klorid/0,5% ecetsav oldattal eluáltuk. After loading the cell culture supernatants onto the chromatography columns, the non-specifically bound proteins were washed with 3 M sodium chloride / 1.5 M glycine, pH 8.9; the antiancrodant antibody activity was eluted with 500 mM sodium chloride / 0.5% acetic acid.
Az antitest szubtípusának meghatározását a fent leírt ELISA technikával analóg módon végeztük, itt azonban a biotinezett antiegér IgG antitest helyett a következő biotinezett szubtípusspecifikus antitesteket alkalmaztuk: antiegér lgG1, antiegér IgM, antiegér IgG2a, antiegér-κ, antiegér lgG2b, antiegér-λ és antiegér lgG3.The antibody subtype was determined in an analogous manner to the ELISA technique described above, but the biotinylated anti-mouse IgG antibody used was the following biotinylated subtype-specific antibody: anti-mouse IgG1, anti-mouse IgM, anti-mouse IgG2a, anti-mouse-Ig, .
Az izolált MÁK 1-2, MÁK 2-29/3 és MÁK 3-27 hibridóma-sejtvonalak antitesttípusának és -szubtípusának (lásd az 1. példát) a vizsgálata mindenkor a következő eredményt adta: lgGI/κ, lgG2b/x és lgGI/κ.The antibody type and subtype of the isolated MAK 1-2, MAK 2-29 / 3 and MAK 3-27 hybridoma cell lines (see Example 1) always yielded the following results: IgGI / κ, IgG2b / x and IgGI / κ.
A monoklonális antitestek affinitáskonstansát különböző, az irodalomból ismert technikákkal állapítottuk meg, például egyensúlyi dialízissel, immunprecipitációval vagy ELISA-val [például: J. H. Peters: „Monoklonale Antikörper, Herstellung und Charakterisierung”, Springer Verlag; A. M. Campbell: „Monoclonal Antibody and Immunosensor Technology”, Elsevier, 11. fejezet (1991)].The affinity constant of the monoclonal antibodies was determined by various techniques known in the art, such as equilibrium dialysis, immunoprecipitation, or ELISA [e.g., J. H. Peters, "Monoclonal Antikörper, Herstellung und Charakterisierung", Springer Verlag; A. M. Campbell, "Monoclonal Antibody and Immunosensor Technology," Elsevier, Chapter 11 (1991)].
ELISA módszer szerint [J. Immunoi. Methods, 77, 305-319 (1985)] a természetes ankroddal szembeni affinitásra a következő értékeket kaptuk (1. táblázat).By ELISA [J. Immunol. Methods, 77, 305-319 (1985)] gave the following values for affinity for the natural anchor (Table 1).

A lemezeket 37 °C-on inkubáltuk, majd humán fibrinogént (1,5 mg) adtunk a lemezekhez. A 37 °C-on való inkubálás után keletkezett fibrint 340 nm-en optikai sűrűség (OD)-méréssel határoztuk meg.Anchor and antibodies (cell culture supernatants and purified antibodies) were plated on BSA-saturated plates at various concentration ratios. The plates were incubated at 37 ° C and human fibrinogen (1.5 mg) was added to the plates. Fibrin formed after incubation at 37 ° C was determined by optical density (OD) measurement at 340 nm.
Increasing amounts of neutralizing antibodies were able to neutralize ankrodactivity, which was manifested in reduced optical density (OD; Table 2).
Különösen hatásosnak mutatkozott a MÁK 1-2 antitest, amely magasabb ankrodkoncentrációknál is teljes neutralizációt eredményezett (2. táblázat, az OD a vak értéknek felel meg). A 2. táblázatban a vak érték minden alkotórészt tartalmaz az ankrodon kívül. A legmagasabb OD-értékeket mindenkor a különféle monoklonális antitestek hozzáadása nélküli különböző ankrodadagokkal (50, 25 és 12,5 ng/ml ankrod) mértük (2. táblázat). The MAK 1-2 antibody was particularly effective and resulted in complete neutralization even at higher anchor concentrations (Table 2, OD corresponds to the blank value). In Table 2, the blank value includes all components except the anchor. The highest OD values were always measured with different doses of anchovies (50, 25 and 12.5 ng / ml) without the addition of various monoclonal antibodies (Table 2).
A MÁK 2-29/3 és MÁK 3-27 antitesteknek hasonlóan jó vagy jobb az affinitásuk az ankrodhoz, mint a MÁK 1-2-nek (1. táblázat), az antitest-antigén kötés azonban csak az ankrodmennyiségekhez viszonyított nagyobb antitestmennyiségek adásánál vezetett az enzimaktivitás neutralizációjához.MAK 2-29 / 3 and MAK 3-27 have similar or better affinity for anchor than MAK 1-2 (Table 1), however, antibody-antigen binding only resulted in the administration of higher amounts of antibody relative to anchor quantities. to neutralize enzyme activity.

Ankrod „in vitro neutralizálása tisztított monoklonális antitestekkel A tisztított monoklonális antitestek in vitro neutralizálóhatékonyságának az értékelése az 50%-os neutralizációs értékek fibrinzavarosság-vizsgálatban való összehasonlításával végezhető el.In Vitro Neutralization of Ankrod with Purified Monoclonal Antibodies The evaluation of the neutralization efficiency of purified monoclonal antibodies in vitro can be accomplished by comparing the 50% neutralization values in the fibrin turbidity assay.
Az 50%-os neutralizációs értéket a következőképpen kapjuk: OD negatív kontroll+(OD pozitív kontroll-OD negatív kontroll)/2;A 50% neutralization value is obtained as follows: OD negative control + (OD positive control-OD negative control) / 2;
Negatív kontroll: nincs ankrodhozzáadás (nincs fibrinképződés);Negative control: no anchoring (no fibrin formation);
Pozitív kontroll: ankrod+fibrinogén (maximális fibrinképződés).Positive control: ankrod + fibrinogen (maximum fibrin formation).
Különböző antitest/ankrod hányadosok mellett változó OD-értékeket kaptunk, amikor is az 50%-os neutralizációs pontot a következő antitestkoncentrációknál értük el:Variable OD values were obtained at different antibody / anchor ratios, whereby the 50% neutralization point was reached at the following antibody concentrations:
Az 50%-os neutralizációhoz szükséges antitestmennyiségek viszonyából levezethető, hogy a választott in vitro feltételek mellett a MÁK 1-2 monoklonális antitest 1 órás előinkubálás mellett legalább kétszer jobb, 2 órás előinkubálás mellett körülbelül négyszer jobb, mint a kecskéből származó poliklonális antitest alapú antidotum. From the ratio of antibody to 50% neutralization, it can be deduced that, under the chosen in vitro conditions, the MAK 1-2 monoclonal antibody is at least twice as good at 1 hour pre-incubation and about 4 times better at 2 hours pre-incubation than goat-derived polyclonal antibody.

A különböző antitestkombinációkat tartalmazó sorozatokat PBS/0,1% Tween 20 oldatban két órán át 23 °C-on inkubáltuk, és a továbbiakban a következőképpen kezeltük:- To a different series of microtiter plates prepared in this manner, 10-10 ng / ml of MÁK 1-2-biotin, MÁK 2-29 / 3-biotin biotinylated monoclonal antibody were added and bound to the anode, followed by another antibody (MÁK 1- 2, MAK 2-29 / 3 or MAK 3-27) (1 μg / ml-1 ng / ml) were added to the series so that each possible antibody combination was tested for their overlapping binding sites. Sequences containing different antibody combinations were incubated in PBS / 0.1% Tween 20 for two hours at 23 ° C and subsequently treated as follows:

- Abszorpciómérés 450 nm-en.- Measurement of absorbance at 450 nm.
No decrease in OD was observed with any of the antibody combinations used, i.e., the various monoclonal antibodies were not displaced upon binding to the anchor. These antibodies bind to different epitopes on the anode molecule. Therefore, several antibodies can interact with the anode at the same time. Therefore, various monoclonal antibodies can be used in combination, if necessary and desired, to rapidly neutralize the effect of the anchor.

Vérmintákat (8 térfogat vér+2 térfogat 0,11 M citrát antikoaguláns) vettünk az artéria carotisból (nyaki verőér) az ankrodinfúzió megkezdése előtt, illetve utána 30 és 60 perc múlva. A citrátos vérből centrifugálással nyertük a plazmát, és a fibrinogéntartalmat a Clauss-féle alvadási módszemel határoztuk meg (standardgörbe-készítés: patkányfibrinogén meghatározott mennyiségeinek hozzáadása fibrinogénmentesített patkányplazmához).Narcotized rats were infused with 10 IU / kg body weight of anode for 30 minutes into the tail vein. 10 minutes after the onset of an anodrinfusion, the various test substances, monoclonal antibodies, polyclonal antibodies or placebo, were administered as a bolus of 1 ml / kg body weight intravenously. Blood samples (8 volumes of blood + 2 volumes of 0.11 M citrate anticoagulant) were taken from the carotid artery (carotid artery) before, 30 and 60 minutes after the start of the anodine infusion. Plasma was obtained from the citrated blood by centrifugation and the fibrinogen content was determined by the Clauss clotting method (standard curve: addition of specific amounts of rat fibrinogen to fibrinogen-depleted rat plasma).

It can be seen that MÁK 1-2 was able to stop a further decrease in fibrinogen levels 30 minutes after the start of an anodrinfusion at a concentration of 1.435 mg / kg body weight. To achieve a similar effect from a goat polyclonal antibody, 8.6 mg / kg body weight was required. A polyclonal based antidote had no effect similar to that of MAK 1-2, i.e. 1.5 mg / kg body weight (see control column in Table 4).

A 60 perc után kapott fibrinogénkoncentrációk alapján azt a következtetést vonhattuk le, hogy a MÁK 1-2 a vizsgált feltételek mellett in vivő mintegy hatszor jobban neutralizál, mint a poliklonális antidotum. A MÁK 1-2 neutralizálóhatása valószínűleg jelentősen nagyobb.Based on the fibrinogen concentrations obtained after 60 minutes, it can be concluded that MAK 1-2 is approximately six times more neutral in vivo than the polyclonal antidote under the conditions tested. The neutralizing effect of MÁK 1-2 is probably significantly greater.
