Postupak dobivanja vodene otopine propolisa se odvija na način da se 2kg suhog ekstrakta propolisa prelije s 10 l demineralizirane vode te se termički vođenim procesom pri temperaturi od do uz kontinuirano ultrazvučno miješanje provodi ekstrakcija propolisa. Proces se provodi kontinuirano 18 h nakon čega slijedi stabilizacija vodene otopine propolisa na 5h. Nakon stabilizacije i filtracije vodena otopina propolisa se koristi u izradi medno-propolisnih sirupa. KVANTIFIKACIJA I STANDARDIZACIJA FLAVONOIDA QUANTIFICATION AND STANDARDIZATION OF FLAVONOIDS
Farmakološki najvažnija komponenta propolisa su flavonoidi. Flavonoidi su biljni pigmenti koji kod biljka imaju zaštitničku ulogu od nametnika i osiguravaju okus i boju voća i povrća. Flavonoidi su odgovorni za biološku aktivnost propolisa odnosno terapeutski učinak propolisa jer imaju antibakterijsko, antivirusno, antioksidativno, protuupalno, antitumorsko djelovanje. Pharmacologically, the most important component of propolis is flavonoids. Flavonoids are plant pigments that protect plants from pests and ensure the taste and color of fruits and vegetables. Flavonoids are responsible for the biological activity of propolis, i.e. the therapeutic effect of propolis because they have antibacterial, antiviral, antioxidant, anti-inflammatory, antitumor effects.
Kvantifikacija Quantification
Dvije najčešće metode određivanja flavonoida su spektofotometrijska metoda (UV/Vis spektrofotometrija) i kromatografska metoda (HPLC). Spektrofotometrijskom metodom određuju se skupine flavonoida (npr. flavoni, flavonoli, flavanoni, flavanonoli) dok se kromatografskom metodom određuju pojedini flavonoidi (npr. galangin, kvercetin, naringenin, apigenin, krizin, pinocembrin). Kroz projekt Farmakol, u suradnji sa Farmaceutsko-biokemijskim fakultetom, vršili smo određivanja ukupnih flavonoida spektrofotometrijskom metodom - UV/Vis spektrofotometrijska metoda. UV/Vis spektrofotometrijska metoda se provodi na principu reakcije odgovarajućeg reagensa s funkcionalnim skupinama flavonoida pri čemu nastaje obojeni produkt. Intenzitet boje ovisi o koncentraciji flavonoida u uzorku, a količina ukupnih flavonoida se određuje pomoću baždarnog pravca. Standardizacija propolisa na ukupne flavonoide započinje ekstrakcijom propolisa u alkoholnoj otopini pri čemu je važno da količina pročišćenog propolisa koja se otapa u alkoholnoj otopini bude što veća (najmanje 100 kg) te da je podrijetlo propolisa iz različitih dijelova Hrvatske. Na taj način dobivaju se alkoholne otopine propolisa odnosno na kraju suhi ekstrakti propolisa vrlo stabilnog sastava ukupnih flavonoida. Predmetni izum se odnosi na sirup, dodan med razrijeđen demineraliziranom vodom, pripravak ljekovitih svojstava u obliku soka pogodnog za oralnu primjenu, pri čemu rečeni sirup sadrži homogenu smjesu vodene otopine suhog ekstrakta propolisa, meda u otapalu prihvatljivom za oralnu primjenu, te da dodatno sadrži ekstrakte ljekovitog bilja. Vodena otopina suhog propolisa je standardizirana na flavonoide i sadrži najmanje flavonoida.
