Abstract

Living cells are isolated from tissue of animal and vegetable origin by subjecting the tissue to the action of an enzyme, such as trypsin, and then, after a desired lapse of time, arresting the enzymatic action by a biochemical reaction in an instantaneous and irreversible fashion by introducing an inhibitor of the enzyme which forms a complex with it having no effect upon the vitality of the cells obtained. Specified inhibitors are ovomucoid, the peptidase inhibitors extracted from the pancreas, parotids or ascaris, and lima or soya beans. The cellular suspensions containing the inhibitor/enzyme complex may be used directly, without centrifuging and washing operations, for cell cultures and for virus cultivation. According to examples: (1) cortical zones of pig's liver cut into pieces, are introduced into a suspension of 0.5 g. trypsin in one litre of Hank's liquid. The enzyme treatment is continued for five minutes at pH 7.2 and at the end of this period there is added a sufficient quantity of ovomucoid to render the whole of the enzyme inactive, viz. ten parts of an ovomucoid solution containing 0.5 g. ovomucoid per 100 ml. Hank's liquid, per 100 parts of trypsin suspension. The yield of living cells was very satisfactory; (2) peptidase inhibitor extracted from ox pancreas replaced the ovomucoid of Example 1; (3) the cells obtained by the process of Example 1 were inoculated, after the addition of a nutritive medium, with aphthous fever virus.
