Abstract

A process for the culture of the human common cold virus by incubation in a tissue culture system comprising a culture medium containing cells of human or monkey origin capable of supporting the growth of the strain or strains of virus selected is characterized in that the culture medium is maintained at a temperature of from 30 DEG to 36 DEG C. and at a pH of between 6.4 and 7.4. The vessel containing the medium and cells is rolled during incubation. A preferred pH is 6.4 to 7.2 and especially 6.8 to 7.2, the preferred temperature being 31 to 35 DEG C. and especially 32 DEG to 34 DEG C. The medium may be Hank's saline medium fortified with an enzymatic hydrolysate of lactalbumin. The medium may contain (a) an alkali metal bicarbonate in a concentration of 0.001 M and 0.01 M or a sodium phosphate buffer having a pH of 7.14 and 7.69 at a concentration of 0.05 M, (b) anti-bacterial and/or antifungal antibiotics, e.g. penicillin, streptomycin and mycostatin. After incubation for 2 to 14 days, the aqueous liquid is separated from the solids and may be used as a vaccine especially after concentration of the antigens, e.g. by ultra-centrifuging, dialysis or ion-exchange. The invention also relates to a method of testing the potency of a common cold virus preparation which comprises carrying out the culturing process of the invention, observing the cell material under a microscope at a time when a substantial number of primary plaques has been formed but before the formation of secondary plaques, counting the number of primary plaques and comparing the number of primary plaques with the number obtained in corresponding manner with a control preparation of virus.
