Abstract
A chromatographic column for use with solvents causing cellulose to swell comprises a rigid core (1) having a paper strip wound thereon to form a reel (2) functioning as the separation medium, and surrounded by a close-fitting flexible sleeve (3) in turn having a paper strip <PICT:0883766/III/1> wound thereon to form a reel (4) surrounded by a close-fitting flexible sleeve (5). In use, the swelling solvent is applied to both reels and the second reel functions to reduce pressure inequalities in the first reel as a result of swelling. The numerals relate to the Figure in which eluted liquid is collected via funnel 6 in vessel 9 and a drip tray 7 is provided for liquid which has passed through the second reel. The rigid core may be made of polyethylene or glass; the sleeves are also preferably made of polyethylene. Paper having ion exchange properties may be employed, e.g. paper comprising one or more ion exchange resins (as in Specification 814,363) or a paper prepared from carboxymethyl cellulose, aminoethyl cellulose, diethylaminoethyl cellulose, 3(N:N diethanolaminoethoxy) 2(hydroxy) propyl cellulose, cellulose citrate, or cellulose phosphate. Examples mention the separation of an aqueous HCl solution of amino-acids using either an aqueous methyl ethyl ketone/propionic acid mixture or an aqueous sodium acetate/HCl buffer solution as developing solvent and the separation of methyl orange and xylene cyanol using water solvent. Aqueous electrolyte/organic solvent developing solutions may also be used.
