Abstract
The invention comprises the tetrahydrate, hydrobromide, hydriodide, nitrate, sulphate, sulphanilate and naphthalene-b -sulphonate of cocarboxylase (aneurin pyrophosphoric ester), and the preparation thereof and of other salts of cocarboxylase by dissolving in water the mixture of orthophosphoric acid and various phosphoric acid esters of aneurin obtained by phosphorylation of aneurin, obtaining from this solution with the aid of one or more suitable ion exchange resins an aqueous solution of cocarboxylase tetrahydrate, isolating this compound in crystalline form therefrom by removal of the solvent water or preferably by addition of a precipitating agent (e.g. a lower alcohol or acetone) and, if desired, converting the crystalline tetrahydrate into a salt of cocarboxylase by treatment with an acid. The solution from which the cocarboxylase tetrahydrate is crystallized may be obtained by adding a precipitating agent to the aqueous solution of the phosphorylation mixture, redissolving and reprecipating two or three times, if desired, and dissolving the resulting syrup in a little water, and then: (a) passing the solution over a weakly basic anion exchange resin and separating the portion off the eluate running off at pH5.5 to 1; or (b) passing the original solution or that obtained according to (a) over a strongly basic anion exchange resin and separating the portion of the eluate running off at pH 5.5 to 1; or (c) passing the solution obtained according to (a) over a cation exchange resin (which retains aneurin orthophosphoric ester and allows a purified solution of cocarboxylase tetrahydrate to run through. Aneurin orthophosphoric ester may be subsequently eluted with 10% hydrochloric acid, and is also obtainable as a by-product in (a) and (b)). Alternatively, an aqueous solution of a cocarboxylase salt, especially one still containing impurities, may be treated with an ion exchange resin, preferably an anion exchange resin.
