Abstract

The invention comprises a 3-keto- D 4,6-steroid compound having the formula I, II or III <FORM:0794392/IV (b)/1> <FORM:0794392/IV (b)/2> <FORM:0794392/IV (b)/3> wherein Y is OH < H or =O, and R is OH or OAcyl (Acyl having up 10 carbon atoms), and a process for the preparation of a 3-keto- D 4,6-steroid compound which comprises reacting a 3-keto- D 4-pregnene with chloranil in an inert solvent at an elevated temperature of at least about 125 DEG C. As solvents there may be used xylene, orthodichlorobenzene, and other inert organic solvents, preferably having a b.p. above 125 DEG C. As starting materials there may be used progesterone, hydrocortisone, cortisone, cortisone esters, hydrocortisone esters, 14-hydroxy - cortisone, 14 - hydroxy - hydrocortisone, desoxycorticosterone, D 14 - hydrocortisone, D 14 - cortisone, 14,15 - epoxidocorticosterone, 14,15 - epoxidohydrocortisone, and esters of these. Examples describe the production of D 6-dehydrohydrocortisone acetate, D 6 - dehydroprogesterone, D 6 - 14 - hydroxyhydrocortisone propionate, D 6 - dehydrocortisone acetate, D 6 - dehydro - 17-hydroxydesoxycorticosterone - diacetate, D 6-dehydrodesoxycorticosterone acetate, D 4,6,14-pregnatriene - 11b ,17a ,21 - triol - 3,20 - dione acetate, D 6,14 - didehydrocortisone acetate, D 6 - dehydro - 14,15 - a - epoxidocortisone acetate, D 6 - dehydro - 14a ,15a - epoxidohydrocortisone acetate, D 6 - dehydro - 14a -hydroxycortisone acetate, and D 6 - dehydro-14a - hydroxy - hydrocortisone acetate.ALSO:The invention comprises a therapeutic composition comprising a compound having the formula I, II or III <FORM:0794392/VI/1> <FORM:0794392/VI/2> <FORM:0794392/VI/3> wherein Y is <FORM:0794392/VI/4> , or = O, and R is OH or OAcyl (acyl having up to 10 carbon atoms), and a pharmaceutical carrier. Administration may be by oral, intramuscular or intra-articular route, or topically in the form of ointments. Injection may be effected in suspension in water or in saline solution.
