Abstract

Tachysterols and esters thereof are obtained by treating a neutral or weakly alkaline solution of precalciferol or an ester thereof with a catalyst known to be capable of converting cisisomers into trans-isomers. Iodine or eosine may be used as catalyst, the conversion being effected under the action of light. As starting compounds there may be used precalciferol-2 or precalciferol-3 or esters thereof. These may be in the form of the product obtained by ultraviolet radiation of a solution of a 3-hydroxy-D 5,7-sterol or an ester thereof from which any unreacted 3-hydroxy-sterol or ester thereof has been removed. Solvents for the reaction are petroleum ether, ligroine, benzene, toluene, cyclohexane, diethyl ether and carbon disulphide. Preferably an acid-binding agent such as pyridine, collidine or quinoline is added. The tachysterol can be isolated by crystallization as an ester of 3,5-dinitro-4-methyl benzoic acid. Dihydrotachysterol is obtained by subjecting tachysterol to reduction with an alkali metal and an aliphatic alcohol, e.g. sodium and propanol, 2-methyl-butanol-2, ethylene glycol or glycerol. Examples describe the preparation of tachysterol-2, dihydrotachysterol-2 and dihydrotachysterol acetate.
