Abstract

The invention comprises benzhydryl ethers of tropine in which either or both of the benzene rings of the benzhydryl variety is/are mono- or di-halogenated by chlorine, bromine or iodine, together with their acid addition salts and quaternary ammonium salts, and methods for their preparation by reaction of (a) a 3-halotropane with a halogenated benzhydrol, (b) tropine with a halogenated diphenyldiazomethane, or (c) tropine with a halogenated benzhydryl halide. In examples: (1) 4-chlorobenzophenone is treated with hydrazine in anhydrous alcohol and the resulting 4-chlorobenzophenone hydrazone oxidized with yellow mercuric oxide or manganese dioxide in petroleum ether to 4-chlorodiphenyldiazomethane, which after filtration is concentrated and immediately reacted with tropine in benzene under reflux, yielding tropine 4-chlorobenzhydryl ether, isolated as the hydrochloride; (2) tropine is reacted with 4-chlorobenzhydryl chloride; and (3) 3-chlorotropane is reacted with 4-chlorobenzhydrol, both methods yielding tropine 4-chlorobenzhydryl ether. Other tropine benzhydryl ethers mentioned are the hydrochlorides of 4-bromo-, 4-iodo-, and 4 : 41-dichloro-, the hydro-iodides of 2-chloro- and 4-chloro-, the hydrobromide, methobromide and methochloride of 4-chloro-, and tropine 2,4-dichlorobenzhydryl ether.
