Abstract
The invention comprises compounds of the formula <FORM:0754162/IV(a)/1> where n is 3 or 4 and R is an alkyl group of 1-4 carbon atoms and their preparation by treating a cyclobutyl alkyl ketone or a cyclopentyl alkyl ketone with sodium acetylide in liquid ammonia. In examples (1) acetylene is passed into liquid ammonia to which metallic sodium is added and the resulting solution of sodium acetylide is treated with cyclobutylmethylketone to give 1-cyclobutyl-1-ethynyl-ethanol which extracted with ether and separated by fractional distillation; and (2) cyclopentylmethylketone is treated with sodium acetylide as in (1) to give 1-cyclopentyl-1-ethynyl ethanol.ALSO:Pharmaceutical preparations having hypnotic activity comprise compound of the general formula <FORM:0754162/VI/1> where n is 3 or 4 and R is an alkyl group of 1 to 4 carbon atoms (see Group IV(b)) in admixture with a non-toxic carrier. The carrier may be a liquid capable of giving solutions, elixirs or emulsions or a solid absorbent such as diatomaceous earth or magnesium oxide, capable of being pressed into tablets. The carbinols may also be incorporated with peanut oil, sesame oil, corn oil or other edible oils and enclosed in capsules. Suppositories are made by incorporating the compounds in a base material such as a solid polyethylene glycol or a mixture of white wax and cocoa butter. Solutions for intraperitoneal administration may also be prepared. Examples of pharmaceutical preparations comprise capsules containing a mixture of polyethylene glycol and 1-cyclobutyl-ethynyl ethanol or 1-cyclopentyl-1-ethynyl ethanol enclosed in soft gelatin and a syrup containing one of these specified carbinols admixed with ethyl alcohol, glycerine, methyl p-hydroxybenzoate, glucose, sucrose, vanilla extract, water and a dye.
