Abstract

A method of culturing a penicillin-producing micro-organism comprises culturing the organism in contact with a liquid nutrient medium including as an assimilable source of carbon and energy a substance which is a member of a group consisting of (1) fats and (2) fatty acids having at least 14 carbon atoms in the molecule, the amount of any other assimilable source of carbon and energy in the medium being substantially less than the total caloric requirement of the culture. The said substance may provide the whole of the assimilable carbon source, or part, the balance being carbohydrates such as lactose and glucose. Specified fats are lard oil, soybean oil, linseed oil, cottonseed oil, peanut oil, coconut oil, corn oil, castor oil, sesame oil, crude palm oil, tallow, sperm oil, olive oil, tristearin, tripalmitin, triolein and trilaurin. Suitable fatty acids are stearic, palmitic, oleic, linoleic and myristic. Comminuted whole seeds especially beans, peanuts cottonseed and linseed may be used. The liquid oils or fatty acids are preferred; the solids may be melted before incorporating in the nutrient medium, or dispersed therein during the sterilization step.
