Abstract

A method of culturing antibiotic producing actinomycetes comprises culturing the actinomycetes in contact with a liquid nutrient medium including as an assimilable source of carbon and energy a substance which is a member of the group consisting of (1) fats and (2) fatty acids having at least 14 carbon atoms in the molecule, the amount of any other assimilable source of carbon and energy in the medium being substantially less than the total caloric requirement of the culture. Thus the carbohydrate content of the culture medium is replaced wholly or in part by such a fat or fatty acid, or mixture of two or more such substances, on a weight basis, or preferably on a caloric basis. The liquids are preferred; the solids are melted before mixing in the culture medium, or may be dispersed therein during the sterilization step. Specified fats are lard oil, soybean oil, linseed oil, cottonseed oil, peanut oil, coconut oil, corn oil, castor oil, sesame oil, palm oil, tallow, sperm oil, olive oil, tristearin, tripalmitin, triolein and trilaurin. Suitable fatty acids are stearic, palmitic, oleic, linoleic and myristic acids. Comminuted whole seeds, e.g. ground, crushed or macerated with water, may be used especially beans, peanuts, cottonseed and linseed. Examples relate to the culturing of (a) streptomyces griscus (streptomycin), streptomyces fradiae (neomycia) and streptomyces aureofaciens (aureomycin). Other microorganisms specified are streptomyces lavendulae (streptothricin) and streptomyces venezuelae (chloromycetin).
