Abstract

A process for the manufacture of orthophosphoric acid esters comprises treating a metallo-derivative of an organic compound of the kind R.OH, wherein R is a substituted or unsubstituted alkyl, cycloalkyl or aryl group and wherein the hydroxyl group is attached to a carbon atom with tetrabenzyl pyrophosphate. If desired, the benzyl groups of the dibenzyl orthophosphate so formed may be removed by catalytic hydrogenation. The substituent may be a fused ring or fused rings. Alkali metal, e.g. sodio derivatives, are the preferred metallo-derivatives. In examples: (1) stillboestrol is reacted with sodium ethoxide to give the sodium derivative and the latter is reacted with tetrabenzyl pyrophosphate to give stilboestrol diphosphate tetrabenzyl ester. Similarly prepared are the dibenzyl orthophosphates of (2) hexoestrol, (3) oestrone, (4) 3 : 41-dihydroxychalkone, (5) ethyl alcohol, (6) dl-a -tocopherol, (7) 2-methylnaphthohydroquinone, (8) d-galactose, (9) 2,3-isopropylidene methyl-d-ribofuranoside, (10) 2,3,4-diisopropylidene-d-sorbose. By hydrogenation using Adams palladium oxide and palladium charcoal catalysts, some of the products are converted to (3) oestrone-3-phosphate, (5) ethyl phosphate and its barium salt, (6) dl-a -tocopherol phosphate and its di-ammonium salt, (7) 2-methylnaphthohydroquinone diphosphate and its di-barium salt, (8) d-galactose-6-phosphate and its barium salt, (10) d-sorbose-1-phosphate and its barium salt. The product of (9) is converted to the barium salt of d-ribose-5-phosphate in the manner described in J.C.S. (1949), 2486.
