Abstract
2-keto-laevo-gulonic acid is prepared from laevo-sorbose by forming the corresponding bis-methylene ether derivatives by treatment with carbonyl compounds, oxidizing them to the corresponding acid in alkaline solution, and then splitting off the carbonyl compounds by heating with water or dilute acids. Suitable carbonyl compounds are formaldehyde, acetone, methyl-ethyl-ketone, and benzaldehyde. Suitable oxidizing agents are permanganate and ferri-cyanide. It is not necessary to isolate the intermediate products. In examples: (1) sorbose is suspended in acetone, sulphuric acid added slowly and the mixture stirred 24 hours, then neutralized with ammonia gas, and filtered. After the acetone is distilled off a viscous oil remains which is crystallized from benzine. The diacetone-laevo-sorbose so produced is dissolved in dilute potash, and potassium permanganate added with stirring, the product filtered, evaporated to dryness in vacuo and extracted with alcohol and the potassium salt of the diacetone compound of 2-keto-laevo-gulonic acid crystallized. The free acid is produced by the action of hydrochloric acid in ice cold solution, and the 2-keto-laevo-gulonic acid isolated by heating on the water bath; (2) 50 kilos benzaldehyde are poured over 2 kilos 1-sorbose and 0,5 kilo hydrochloric acid gas passed in. Ether is added, the solution washed with alkali, dried, and oxidized with alkaline permanganate. The di - benzal - 2 - keto - laevo-gulonic acid is liberated by mineral acid from an ether solution of the reaction product, and the 2-keto-laevo-gulonic acid itself formed by boiling with alcoholic hydrychloric acid.
