Abstract
A malodour-blocking perfume composition comprising one or more ingredients selected from cyclic nucleotide-gated ion (CNGA2) channel blocking agents. The present invention has the capability of decreasing the flux of calcium ions in the CNGA2 channels, thereby decreasing the activation of the olfactory sensory neurones. The ingredients may be selected from three groups having either a high calcium flux reduction, a moderate calcium flux reduction or a low calcium flux reduction. The perfume composition may be used in consumer products such as household surface cleaning products, fabric treating products, air-freshening products, oral care products or a product for treating human body odour.
Description
Improvements in or relating to organic compounds This disclosure relates to perfume compositions comprising cyclic nucleotide-gated ion channel (so-called CNGA2 channel) blocking agents and being especially useful in counteracting malodours, and in particular ammonia and sulphur malodour. More particularly, this disclosure relates to perfume composition comprising perfume ingredients having the capability of decreasing the flux of calcium ions in said channels, while still being perceivable as odourants.
Malodours are offensive, unpleasant odours emitted into an atmosphere from a source of malodour. Typical sources include fabrics, hard surfaces, skin, and hair. Malodours have either personal or environmental origin. For example sweat, urine and feces malodours are personal in origin, whereas kitchen and cooking malodours as well as chemical compounds (organic and inorganic chemical compounds) are of environmental origin.
Amines, thiols, sulfides, short chain aliphatic and olefinic acids, e.g. fatty acids, are typical of the chemicals found in and contributed to sweat, household, and environmental malodours. These types of malodours typically include indole, skatole, and methanethiol found in toilet and animal odours; piperidine and morpholine found in urine; pyridine and triethyl amine found in kitchen and garbage odours; and short chain fatty acids, such as 3-methyl-3-hydroxyhexanoic acid, 3-methylhexanoic acid or 3-methyl-2-hexenoic acid, found in axilla malodours.
Malodours chemical compounds also found in industry, in particular chemical industry, for example, in production sites. The malodour may be originated from compounds indispensable as starting material, or formed as intermediates in a chemical process. For example organic sulfides such as DMDS (dimethyl disulfide) are useful additives for steam cracking, but unfortunately do posses a very unpleasant odour which makes its use difficult. Further, non-limiting examples of chemical compounds possessing an unpleasant odour are methyl mercaptan, iso-valeric acid (3-methylbutanoic acid), dimethyl trisulfide, and dimethyl sulfide. Whereas these highly malodorous chemical compounds are essential in some areas, they are unpleasant to handle due to their offensive malodour even when emitted in very low concentrations into an atmosphere.
Fragrance in an atmosphere or sprayed on to a substrate (including skin) is often used to counteract malodours in a number of situations. Over the centuries, perfumers have developed the art of achieving the proper malodour counteracting effect by combining powerful odorants having a pleasant smell that overwhelms the malodour. Perfumers have also learnt how to use malodours in the perfumed composition in such a way that the overall impression given to the consumer is that of a nice and pleasant smell. Other ways to suppress malodours encompass mixing with an ingredient that causes a negative deviation according to Raoult's law, elimination of the malodour by absorption of the malodour by a porous or cage-like structure, and avoidance of the formation of malodours by such routes as antimicrobials and enzyme inhibitors. Although the methods known in the art have the ability to neutralize certain malodours, there still remains a need for further compounds which are even more efficient against malodours.
Patent Application W02014064101 discloses the use of essentially odourless compounds that unexpectedly reduce the perception when applied in such a concentration that the ratio between the compound of formula (I), or a mixture thereof (malodour blocker), and malodour is from about 1:1 (mol weight) to about 1:100 (mol weight), e.g. between 1:10 to 1:70 (such as 1:30 or 1:50), in the atmosphere. It is suggested that such compounds do block the cyclic nucleotide-gate ion channel and thus reducing the number of calcium ions that enter the cell and thereby decreasing the activation of the olfactory sensory neurons.
Whereas the skilled person will appreciate that essentially odourless compounds may have such an effect on the olfactory sensory neurons, the applicant has found that this effect can also be produced by an odorant molecule, provided a sufficient level of this molecule is provided in the atmosphere by any conventional fragrance disseminating methods, such as evaporation, aerosol spray and the like.
Furthermore, the applicant has found that in vitro bioassay using heterologously expressed CNGA2 homotetrameric channels, which, according to Nache et al. (Nache, Schulz, Zimmer, Kusch, Koopmann, Hagen & Benndorf. J. Physiol (2005) 569, 91-102), hereby incorporated by reference, are activated with the same kinetics as the native heterotetrameric olfactory channels, provide a suitable method to identify perfumery ingredients having CNGA2 channel blocking ability. Upon activation with dibutyryl-cyclic AMP (db-cAMP) the channel opens and calcium flows into the cells.
The intracellular concentration of calcium is measured on the FlexStation using a calcium-sensitive fluorescent marker. Following activation with db-cAMP the test compound is applied to the activated cells to study whether it had an effect on the calcium flow through the channel. A channel blocker does result in a reduction of the calcium influx and its effect is dose dependent.
Hence, the method provides furthermore dose-response information that is useful for determining the optimal level of CNGA2 channel blocking ingredients in the perfume composition. On the other hand, using such in vitro assessment method, perfumery ingredients can be classified according to their CNGA2 channel blocking activity) by measuring the extent of calcium ions flux depression in the CNGA2 channel, using conventional fluorescence assessment techniques. The perfumery ingredient can therefore be classified Class A ingredients having high CNGA2 channel blocking activity, Class B ingredients having moderate CNGA2 channel blocking activity and Class C ingredients having zero CNGAZ channel blocking activity.
Hence, in a first embodiment of the present invention it is provided a series of perfumery ingredients belonging to class A, B and C, according to the above definitions, whereas Class A ingredients have a calcium flux reduction index of more than 4, on a scale 5, wherein 5 is the maximal calcium flux reduction observed in the data set investigated in this work, class B ingredients have a calcium flux reduction index from 1 to 3 on the same scale and class C ingredients have no effect on the calcium flux (score 0 on the same scale).

In an embodiment of the present invention there is provided a malodour-blocking perfume composition comprising one or more group A compounds, one or more group B compounds, optionally one or more group C compounds, or mixtures thereof. Individual group A or group B ingredients may be present in amounts of 0.01 to 100% of the perfume composition.
S In an embodiment of the present invention is provided a perfume composition comprising more than 25% by weight of Group A ingredients, more particularly more than 35% and still more particularly more than 50% by weight of Group A ingredients.
In an embodiment of the present invention is provided a perfume composition comprising more than 25%, more particularly more than 35% by weight of GROUP A ingredient and more than 35%, still more particularly 45%, and still more particularly 50% by weight of Group B ingredients.
Aside from the aforementioned perfume ingredients, a perfume composition of the present invention may contain other non-functional perfume ingredients, which are traditional perfume ingredients employed for the fragrance or hedonic benefits. Reference to suitable non-functional perfume ingredients can be found in any number of text books including the book by S.Arctander, Perfume and Flavor Chemicals, 1969, Montclair New Jersey, or any of its more recent editions.
Perfume compositions of the present invention may contain other non-perfume ingredients that are commonly employed in said compositions.
In another aspect of the present invention there is provided a consumer product comprising said perfume composition.
The amount of the malodour-blocking perfume compositions that may be employed in said consumer product should be an amount effective amount that provides an agreeable perfume effect, whilst abating a given malodour. The exact amount employed may vary depending upon the type of malodour blocking perfume ingredients employed and the level of malodour blocking desired. In general the group A, B, and optionally C ingredients (or mixtures thereof) may be employed in amounts ranging from 0.1 to 100%, more preferably 0.5 to 80% based on the weight of the consumer product.
The present invention encompasses consumer products for treating/cleaning household surfaces, including countertops, cabinets, walls, floors, bathroom surfaces, and kitchen surfaces.
The present invention also encompasses the method of dispersing an effective amount of the perfume composition for reducing malodour onto said household surfaces.
The present invention encompasses consumer products for treating fabrics or fabric articles, for example by applying a mist or a spray of said perfume composition onto said fabric or fabric articles. The fabric and/or fabric articles include, but are not limited to, clothes, curtains, drapes, upholstered furniture, carpeting, bed linens, bath linens, tablecloths, sleeping bags, tents, car S interior, e.g., car carpet, fabric car seats, shower curtains, and the like.
The present invention also extends to a method of dispersing an effective amount of the perfume composition for reducing malodour onto said fabric and/or fabric articles.
The present invention relates to consumer products for treating malodour impressions on or in rubbish or recycling receptacles.
The present invention also extends to a method of dispersing an effective amount of the perfume composition for reducing malodour impression on or in rubbish or recycling receptacles.
The present invention relates to consumer products for treating malodour impressions in the air.
Consumer products include all manner of air-freshening devices including sprays, candles, oils, gels, beads, and plug-ins.
The present invention also extends to a method of dispersing an effective amount of the composition for reducing malodour impression into the air to neutralize malodour.
The present invention relates to consumer products for treating malodour impressions of animals including cat litter, pet bedding and the like.
The present invention also extends to a method of dispersing an effective amount of the composition for reducing malodour impression on cat litter, pet bedding and the like.
The present invention relates to consumer products for treating malodour impressions in the oral cavity, including hard and soft confections, mouth washes, tooth pastes, denture cleaners, dental floss, and the like.
The present invention also extends to a method of dispersing an effective amount of the composition for reducing malodour impression in the oral cavity.
The present invention relates to consumer products for treating human malodour, in particular body or hair malodour, and still more particularly axilla malodour.
The present invention also extends to a method of dispersing an effective amount of the perfume composition for reducing human malodour impression.

Example 1
S Sensory validation A mixture of five Group A ingredients comprising 0.5% by weight (1-methyl-2-(1R,3R)-2,2,3- trimethylcyclopentyl)methyl)cyclopropyl)methanol, 1% by weight (E)-3,7,11-trimethyldodeca- 1,6,lotrien 301, 1% by weight 1-(2,2,6-trimethylcyclohexyl)hexan-3-ol, 1% by weight (E)-3,7,11-trimethyldodeca-6,1O-dien-1-ol and (E)-3,7,11-trimethyldodeca-6,10-dienal in diethyl phtalate. A second mixture of five Goup C ingredients comprising 0.5% by weight cinnamyl acetate) 0.5% by weight (E)-1,2-dimethoxy-4-(prop-1-en-1-yl)benzene, 0.5% by weight 4,4,8- trimethyltricyclo[6.3. 1.02,5jdodecan-1-yl acetate and 0.8% by weight 79-dimethylspiro[5.5]undecan-3-one in diethyl phthalate. The concentrations were chosen in such a way that the odour of both mixtures was moderate and not overpowering. Isovaleric acid was used as reference nialodour at a concentration of 0.005% by weight and was tested against 2.5% by weight of above mixtures on blotters. The assessors smelled first the reference sample with a low isovaleric acid concentration of 0.0005% by weight in diethyl phytalate and then a second reference sample with high isovaleric acid concentration of 0.005% by weight in diethyl phatalate, in order to provide a normalized answer. 5 panellists of 8 did report a reduction in the perceived isovaleric acid intensity when the mixture with Group A ingredients was used whereas no panellist was able to perceive any reduction of perceived isovaleric acid intensity when the mixture of Group C perfumery ingredients was used, nothwithstanding that both group C and group A compounds are perfume ingredients. It was concluded that the reduction of malodour perception was due to CNGA2 channel blocking effects more than to odour masking effects of perfume ingredients.

- 2. A perfume composition according to claim 1, wherein each group A or group B ingredient may be present in an amount of between 0.01 and 100%.
- 3. A perfume composition according to claim 1 comprising more than 25% by weight of Group A ingredients.
- 4. A perfume composition according to claim ito claim 3 comprising more than 35% by weight of Group B ingredients.
- 5. A consumer product comprising 0.1 to 100% by weight of a perfume composition as defined in any of the claims ito 4.
- 6. Use of a perfume composition as defined in any of the claims ito 4 as a malodour-blocker in a consumer product composition.
- 7. Use according to claim 6, wherein the consumer product composition is selected from a household surface cleaning product, a fabric treating product, an air-freshening product, an oral care product, or a product for treating human body odour.
