Abstract

1526205 Stabilizing interferon G D SEARLE & CO Ltd 31 Oct 1975 [7 Nov 1974] 48135/74 Heading A5B Aqueous interferon preparations are stabilized against inactivation due to mechanical stress, (i.e. the biological inactivation caused by conditions encountered during purification, concentration, packaging procedures, filtration and storage, probably due to formation of sulphydryl group interlinkage or dissulphide interchange reactions to form inter- or intra-molecular disulphide bridges), by treatment with an agent which will selectively retain interferon sulphydryl groups in the reduced state but not reduce essential disulphide linkages, or an agent which will selectively block interferon sulphydryl groups. Suitable agents retaining sulphydryl groups in a reduced state are reduced glutathione, thiodiglycol, thioethanolamine, C 1-7 thioalkanoic acids, monothioglycerol, dithiotreietol, thioctic acid, cysteine, N-acetylcysteine and N- acetyl homocysteine. These materials may subsequently be removed from the interferon solution by dialysis against a buffer solution. Agents which block the interferon sulphydryl groups are pchloromercuribenzoate and phydroxymercuribenzoate, which cannot be removed by dialysis. The interferon preparation may be produced from a fibroblast cell culture.
