Abstract

1513746 Determining thioamides JOHN WYETH & BRO Ltd 24 May 1976 [27 May 1975] 23208/75 Heading G1N A primary thioamide (R-CSNH 2 ) is determined by potentiometric titration with a standard solution of silver nitrate using a metallic silver indicator electrode. The reference electrode may be calomel or glass and both reagents are preferably in an aqueous organic solvent; the thioamide in aqueous ethanol and the silver nitrate in aqueous isopropanol. When the thioamide is present as its hydrochloride, and silver nitrate solution is slowly added to it, the potential curve shows two end points corresponding to the thioamide and to the hydrochloride. When the thioamide is present in basic form, it is preferably added to the silver nitrate solution. Thioamides can be determined by this method in the presence of normal pharmaceutical carriers. A glass reference electrode is necessary when the carrier is dibasic calcium phosphate.
