Abstract

1500377 Cephalosporin - derived thiocarbamates and thiocarbonates E R SQUIBB & SONS Inc 4 April 1975 [10 May 1974 (2) 17 May 1974] 13938/75 Headings C2C and C2J [Also in Division C3] The invention comprises compounds of formula wherein A is O, S or NH, R is H, C 1-7 alkyl, phenyl-C 1-7 -alkyl, tri-(C 1-7 alkyl)stannyl, a salt forming ion or the group and in addition, phenyl when A is O or S; R 1 is H, C 1-7 alkyl, phenyl, thienyl, furyl and in addition, C 5 -C 6 cycloalkyl and C 5 -C 6 cycloalkadienyl when A is S; and pyridyl when A is O; R 2 is C 1-7 alkyl, phenyl-C 1-7 -alkyl when A is O or NH and C 1-7 alkoxy methyl or phenyl when A is NH; R 3 is H, OH or C 2-8 alkanoyloxy; and R 4 is C 1-7 alkyl, phenyl or phenyl C 1-7 alkyl. In examples, these compounds are prepared by (1) N-acylating the corresponding 7-aminocephem compounds, or (2) converting R= CHPh 2 to H using CF 3 CO 2 H. Starting materials prepared have the formulµ HSCHPhCO 2 CHPh 2 and R 2 -A-C(:O)SCHR 1 CO 2 H, and include also an acid chloride, benzhydryl ester and 1- hydroxysuccinimide ester of the latter. Therapeutic or disinfectant compositions having antibacterial activity comprise compounds of the above formula. The therapeutic compositions may be administered orally or parenterally.
