Abstract
1491261 Biochemical hormone production COMMUNITY BLOOD COUNCIL OF GREATER NEW YORK 16 May 1975 [17 May 1974] 20967/75 Heading C2U [Also in Divisions A5 C3 and C6] A method of biochemical transformation or synthesis involves, according to the preferred procedure, passing a metabolic precursor over or through hollow porous fibres into contact with mammalian cells (specified) disposed on the internal or external wall respectively, of the fibres, contacting this wall of the fibres with a physiologically compatible aqueous medium, and collecting the resulting metabolite or metabolic product. Example 3 describes the transformation of a metabolic precursor #-5- pregnenolone into the metabolic product corticosterone with 32% yield by passage of a cell culture medium containing the #-5-pregnenolone dissolved in ethanol through some 400 fibres having a coating of rat adrenal cortical cells disposed on the exterior walls. The fibres were made by depositing collagen solution on to preformed hollow filaments of polytetrafluoroethylene and cutting the coated filaments into 5 cm. lengths. They were porous to molecules of M.W. up to 1000. Similarly testosterone and progesterone are obtained from Eagle's medium supplemented with amino acids and fetal calf serum by contact with testicular hormone producing cells (interstitial cells of Leydig) and ovarian hormone producing cells (follicular and corpus luteal cells) respectively.
