Abstract
1471262 Growing viruses C VET Ltd 25 March 1974 [9 Jan 1973] 1199/73 Heading C6F [Also in Division A5] Feline panleucopenia or mink enteritis viruses are grown in a nutrient medium comprising a mink tissue cell strain which contains actively dividing cells. The viruses may be inactivated or attenuated by serial passage in the cell strain to produce antigenic material, which may be incorporated in a vaccine. The cell strain may be of lung, kidney or heart of an adult, or preferably, foetal mink. The cells are rendered aseptic and disaggregated mechanically or enzymatically and cultured by serial passages in a nutrient medium until a diploid or heteroploid cell strain free from extraneous viruses, is produced and the cell strain is seeded with the virus either before or after mixing with nutrient medium. Where the nutrient is added after seeding, the culture is maintained at 20-40‹C for 0À5-2 hours before adding nutrient which is added at a rate of 10-50 ml/10<SP>6</SP> viable cells, after which incubation at 30-40‹C is carried out for 5-7 days. Harvesting is achieved by centrifuging to recover extracellular virus followed by treatment to break down all walls and centrifugation to recover all associated viruses.
