Abstract
1470955 Polymers bearing imide groups; modified nylons NATIONAL RESEARCH DEVELOPMENT CORP 26 March 1974 [4 April 1973] 16148/73 Headings C3A C3H and C3R A water insoluble organic polymer carries substituent groups of the formula where R represents an aliphatic or aromatic hydrocarbon group and X represents a radical capable of reacting with a biologically active material to link it covalently to the polymer. The preferred polymers are aminoethyl cellulose and nylon. The amino groups present on the former and provided on the latter by partial peptide bond cleavage are reacted with ethyl adipimidate in the presence of N-ethyl morpholine to give the substituted polymer, e.g. by acid hydrolysis or treatment with N,N-dimethylaminopropylamine. The substituted polymers are coupled to various enzymes and co-factors by incubation with them in a basic medium. The amino-ethyl cellulose is coupled to lactate dehydrogenase, liver alcohol dehydrogenase, pig heart lactate dehydrogenase, rabbit muscle dehydrogenase, yeast alcohol dehydrogenase, chymotrypsin, glutamate oxalacetate transaminase and NADH peroxidase. The nylon is linked to trypsin and rabbit muscle lactate dehydrogenase.
