1470074 Immunological reagent BLOCK ENG Inc 22 May 1974 [30 July 1973] 22791/74 Heading GIB [Also in Division C3] A radiant energy tagging material which may be used to tag antibody protein molecules by reaction with the reactive end groups is obtained from polymeric backbone molecules with reactive end groups and repeating units having reactive sites of different chemical reactivity, by bonding radicals derived from radiant energy emitting molecules, e.g. fluorescent dyes, to the polymeric backbone molecules by reaction with the reactive sites. The tagged antibody protein material may be used in immunochemistry to detect and classify viruses by antibody-antigen reaction and measurement of radiant energy emission, e.g. fluorescence. Specific polymeric backbone material described is a polyethylene imine with an amine group at one end and a methyl group at the other; the amine end group is blocked by reaction with benzaldehyde and the polymer reacted first with lithium and then with a fluorochrome dye to bind radicals derived from the dye to nitrogen atoms in the repeating units of the polymer by a demetallation reaction; the amine end group is then freed by hydrolysis and converted to a thiocyanate group, by reaction with thiophosgene, which in turn can react to bind the polymer molecule to an antibody protein molecule. Other polymeric starting materials mentioned are polyethylene oxide, polyamides and polymeric carboxylic and amino acids.
