Abstract

1444205 Tetrahydroquinolines and their cyclopenta- and cyclohepta-analogues; organoalkali metal compounds JOHN WYETH & BROS Ltd 23 Jan 1975 [4 Feb 1974] 04950/74 Addition to 1432378 Headings C2C and C2J The invention comprises compounds of formula wherein (a) R<SP>17</SP> is straight-chain alkyl, possibly aryl-substituted and X<SP>1</SP> is CN, CO 2 alkyl, CO 2 aralkyl or CO 2 aryl, or (b) R<SP>17</SP> is Na, K or Li and X<SP>1</SP> is CN, CO 2 alkyl, CO 2 aralkyl or CO 2 aryl, or (c) R<SP>17</SP> is straight-chain alkyl, possibly aryl-substituted and X<SP>1</SP> is CSNHZ where Z is H or C 1-6 alkyl; and in II(a), II(b) and II(c), R<SP>1</SP>, R<SP>2</SP> and R<SP>6</SP> are each H, CF 3 , or an alkyl, aralkyl or aryl radical, any of which radicals may be substituted by alkyl, alkoxy, halogen, NO 2 or CF 3 or R<SP>1</SP> and R<SP>2</SP> taken together represent an alkylene chain -CH 2 (CH 2 ) n CH 2 - wherein n is 1, 2 or 3, R<SP>7</SP> represents H or single or multiple substitution by alkyl, aralkyl or aryl radicals any of which radicals may be substituted by alkyl, alkoxy, halogen, NO 2 or CF 3 and when R<SP>1</SP> and R<SP>2</SP> taken together form an alkylene chain the resulting ring may be substituted by one or more R<SP>7</SP> radicals as defined above, any of the ester radicals X<SP>1</SP> may be substituted by alkyl alkoxy, halogen, NO 2 or CF 3 , and m is 1, 2 or 3, provided that when R<SP>1</SP>-R<SP>2</SP> represents an alkylene chain, then n=m; and the acid addition salts of II(a) and II(c). These compounds are prepared by reacting a starting material of Formula II, but in which R<SP>17</SP> is H and X<SP>1</SP> is as defined except CSNHZ, with M-R<SP>0</SP> (where M is Na, K or Li and R‹ is alkyl, aryl or aralkyl) or with the alkali metal alkoxide or hydride, to give II(b), and alkylating the latter to give II(a). In an example, II(c) is obtained from II(a) (X<SP>1</SP> = CN) by treatment with H 2 S/ pyridine/Et 3 N. Therapeutic compositions having antiulcer activity comprise compounds of Formula II(c), as defined above. The possibility of compounds II(a) (X<SP>1</SP> = CN) having antiulcer activity is also mentioned.
