Abstract

1437783 Substituted (diphenylyl)acetic acids and derivatives BOOTS CO Ltd 3 Sept 1973 [22 Sept 1972] 44013/72 Heading C2C Novel compounds of the Formula I wherein R 1 , R 2 and R 3 are the same or different and are fluorine, chlorine or bromine, Y is COOH, CONH 2 , CH 2 OH or CONHOH, pharmaceutically acceptable esters of the compounds in which Y is COOH and pharmaceutically acceptable salts of the compounds in which Y is COOH or CONHOH may be prepared by (1) reacting the corresponding 4-acetyl-biphenyl with sulphur and morpholine in a Willergerodt reaction to give the thiomorpholide which is hydrolysed to the free acid, (2) esterification of a free acid, (3) hydrolysis of an ester, (4) hydrolysis of an amide (5) hydrolysis of a 4,4-dimethyl- 2-oxazoline ring to a free carboxylic acid group, (6) reduction of an ester group to CH 2 OH, (7) reacting an acid chloride with ammonia or hydroxylamine to form Y=CONH 2 and CONHOH respectively, (8) salt formation and numerous ether standard methods. 4 - Acetyl - 2,2<SP>1</SP>4<SP>1</SP>- trifluorobiphenyl is prepared by forming the fluoroborate of 4-acetyl-2- amino - 21,41 - difluorobiphenyl followed by decomposition thereof in situ. 4-Acetyl-2- chloro - 21,41 - difluorobiphenyl is prepared by subjecting the same starting material to treatment with cuprous chloride. 4 - Acetyl - 2 - amino - 2<SP>1</SP>,4<SP>1</SP> - difluorophenyl is prepared by an Ullmann coupling of 2,4-difluoroiodobenzene and 4 - bromo - 3 - nitroacetophenone to yield 4-acetyl-2<SP>1</SP>,4<SP>1</SP>-difluoro-2- nitrobiphenyl which is reduced to the required compound. Pharmaceutical compositions of the compounds I with the usual excipients show anti-imflammatory, fibrinolytic and thrombolytic activity and inhibit platelet aggregation when administered orally, rectally, topically or parenterally.
