1434099 Hexahydro-1,3-diazocines JOHN WYETH & BRO Ltd 17 May 1974 [31 May 1973] 26079/73 Addition to 1403732 Heading C2C Novel hexahydro-1,3-diazocines of the general formula wherein Ph is a phenyl group optionally substituted by one or more hydroxyl, C 1-6 alkyl, C 1-6 alkoxy, halo-C 1-6 alkyl, halogen, amino or mono- or di-C 1-6 alkylamino radical, R is a hydrogen atom or a phenyl group optionally substituted by one or more hydroxyl, C 1-6 alkyl, C 1-6 alkoxy, halo-C 1-6 alkyl, halogen, amino or mono- or di-C 1-6 alkylamino radical and R<SP>1</SP> is a hydroxyl group or R and R<SP>1</SP> together form an oxo group and each of R<SP>2</SP> and R<SP>3</SP> is a hydrogen atom or C 1-6 alkyl group, and acid addition salts thereof are prepared (a) when R and R<SP>1</SP> together form an oxo group, by oxidizing the corresponding compound in which R is a hydrogen atom and R<SP>1</SP> is a hydroxyl group; (6) when R<SP>1</SP> is a hydroxyl group and R is other than a hydrogen atom, by reacting the product of (a) with a suitable organo-metallic compound; (c) when R<SP>1</SP> and R are as in (b), by oxidizing the corresponding compound in which R<SP>1</SP> is a hydrogen atom, and (d) when R is a hydrogen atom and R<SP>1</SP> is a hydroxyl group, by reacting a diamine of the general formula with an imino ether of the general formula wherein OR<SP>5</SP> is an etherified hydroxyl group, or an amidine of the general formula followed optionally by salification of the product. Pharmaceutical compositions having hypoglycaemic activity comprise, as active ingredient, a hexahydro-1,3-diazocine of the first general formula above, wherein R<SP>1</SP> is a hydroxyl group and R is other than a hydrogen atom, or a pharmaceutically acceptable acid addition salt thereof, in association with a pharmaceutically acceptable carrier.
