1430777 Pyrido[2,3-b]indole derivatives GRUPPO LEPETIT SpA 22 Aug 1974 [10 Sept 1973] 42368/73 Heading C2C The invention comprises compounds of the general Formula I wherein the dotted lines represent an additional bond localized in one of the two possible positions; R is alkyl of 1 to 4 carbon atoms or benzyl; R 1 is hydrogen, alkyl of 1 to 4 carbon atoms, benzyl or aliphatic acyl of 1 to 4 carbon atoms and is present only if the symbol R 2 is absent and the double bond is located between the carbon atoms in position 2 and 3 of the indole ring and R 2 is hydrogen, alkyl of 1 to 4 carbon atoms or benzyl and is present only if the symbol R 1 is absent and the double bond is located between the positions 1 and 2 of the pyridine ring. The compounds of Formula I may be prepared by heating at a temperature between 60‹ and 160‹ C. an indole derivative of the formula or the corresponding acid salt wherein the dotted lines represent a double bond localized in one of the two possible positions; R is C 1 to C 4 alkyl or benzyl, R 1 is hydrogen, C 1 to C 4 alkyl, benzyl or C 1 to C 4 aliphatic acyl if the symbol R 2 is absent and the double bond is located between the carbon atoms in position 2 and 3 of the indole ring; R2 is hydrogen, C 1 to C 4 alkyl or benzyl and is present only if the symbol R 1 is absent and the double bond is located between the iminic substituent and the carbon atom in the position 2 of the indole ring and R 3 is hydrogen or C 1 to C 4 alkyl in an organic polar solvent in the presence of a basic agent and, optionally when in the obtained compound of Formula I either R 1 or R 2 represent hydrogen, substituting hydrogen with C 1 to C 4 alkyl, benzyl or when R 1 in the obtained compound of Formula I is hydrogen also C 1 to C 4 aliphatic acyl, by reaction with an agent selected from C 1 to C 4 alkyl halogenides and benzyl halogenides in dimethyl formamide in the presence of sodium hydride and C 1 to C 4 aliphatic acyl halogenides or anhydrides in the presence of pyridine. The compounds of Formula I are used as pharmaceuticals.
