1427097 Optically active morpholine derivatives IMPERIAL CHEMICAL INDUSTRIES Ltd 22 Aug 1974 [18 Oct 1973] 48490/73 Addition to 1138405 Heading C2C The invention comprises optically-active morpholine derivatives of the general Formula I wherein R<SP>1</SP> stands for hydrogen or a methyl, isopropyl or allyl radical, wherein R<SP>2</SP>, R<SP>3</SP> and R<SP>4</SP> which may be the same or different, stand for hydrogen or a methyl radical, and wherein X stands for a phenyl radical which may be unsubstituted or substituted by one substituent selected from methyl, n-propyl, allyl, propenyl, methoxy, ethoxy, n-propoxy, isopropoxy, nheptyloxy, alkyloxy, phenoxy, phenyl, methylthio and acetyl radicals and a chlorine atom or substituted by two methyl radicals; and the acid addition salts thereof, the optically-active derivative being that enantiomer having the (S) absolute configuration, as hereinafter defined, about the carbon atom of the morpholine ring bearing the side-chain. By a morpholine derivative having the (S) absolute configuration is meant one which, as a solution of its hydrochloride salt in methanol, exhibits a plain negative optical rotatory dispension curve between 16,000 and 43,000 cm.<SP>-1</SP> (6250 and 2330 Š units). The compounds of Formula I may be prepared by (a) resolution of the corresponding racemic morpholine derivative by conventional means, (b) reducing of an opticallyactive compound of the formula with a complex metal halide, (c) for those compounds in which R<SP>1</SP> is hydrogen, removal of the removable α-aryl alkyl or alkyl radical from an optically-active compound of the formula wherein R<SP>5</SP> stands for a removable α-aryl alkyl radical of up to 11 carbon atoms or a removable alkyl radical of 1 to 6 carbon atoms. (d) cyclization of an optically-active compound of the formula wherein Y stands for a displaceable radical, with a base or (e) reacting an optically-active compound of formula wherein Z stands for a displaceable radical, with a phenol of the formula The compounds of Formula I are used as pharmaceuticals.
