1425706 Piperidine derivatives JOHN WYETH & BRO Ltd 28 June 1973 [30 June 1972] 30636/72 Heading C2C The invention comprises piperidine derivatives of the Formula (I): or an acid addition salt thereof, in which A is a C 2 -C 6 alkyl group optionally substituted by a C 1 -C 6 alkoxy group, hydroxy, amino optionally substituted by one or two C 1 -C 6 alkyl groups, halogen, a cyano group, an optionally substituted carbamoyl group of formula (in which X and Y are hydrogen or lower alkyl), a group of formula -COT (in which T is C 1 -C 6 alkyl), or an oxime, an optionally substituted hydrazone, guanidone, aminoguanidone or semicarbazone derivative of the group -COT; or A represents a group of formula -COOB in which B is an optionally substituted C 1 -C 6 alkyl group; and R represents a C 5 -C 7 oycloalkyl group or an aryl or heteroaryl group; with the proviso that when A is an unsubstituted C 2 -C 6 alkyl group and the group RCONH- is in the 4-position of the piperidine ring then R is a phenyl group substituted by one or more substituents at least one of which is in the 2- or 3- position. They may be prepared by (a) reacting a compound of Formula II: with a compound of formula RCOOH or a functional derivative thereof; (b) reacting a compound of Formula III: with an alcohol of formula BOH; (c) reacting a compound having the Formula IV: or an acid addition salt thereof with an alkylating agent AZ where Z is a leaving group or atom ; (d) reacting a compound having an ion of Formula V: or (e) reacting a compound of the Formula VI: in which A<SP>1</SP> is a C 2 -C 6 alkyl group monosubstituted by a carboxyl group or a derivative thereof is amidated to produce the compound where A is an alkyl group monosubstituted by a substituted or unsubstituted carbamoyl group where X and Y are hydrogen or C 1 -C 6 alkyl, and, if desired, converting the free base into an acid addition salt or vice versa. The products may be used as the active ingredients in pharmaceutical compositions.
