1425560 4 - Benzamido - piperidine derivatives JOHN WYETH & BRO Ltd 29 Aug 1973 [1 Sept 1972] 40618/72 Addition to 1345872 Heading C2C Novel compounds III: in which Z is carbonyl, hydrogen and hydroxy or two hydrogen atoms and A is alkylene optionally substituted by a methyl group and containing up to 5 C atoms and their acid addition and quaternary ammonium salts are prepared by reacting 4-benzamidopiperidine with a compound V: in which Y is halogen or tosyl. Compounds I (Z=OH+H) are also prepared by reduction of a compound I in which Z is carbonyl preferably using sodium borohydride. Compounds I prepared are those in which CZ-A is 4-(oxo or hydroxy)-butyl and 2-(oxo or hydroxy)-1- methyl-ethyl. Compounds I have anti-histamine activity and possess α-adrenoceptor antagonism and form with a carrier a pharmaceutical composition which may be administered orally or parenterally.
