Abstract
1419969 Immunostimulating agent INSTITUT PASTEUR 21 Dec 1972 [24 Dec 1971] 59164/72 Heading A5B A pharmaceutical composition comprises (a) an immunostimulating agent obtained by: (i) extracting cells selected from splenic cells, Ehrlich ascite cells, bacterial cells and Saccharomyces cerevisiae, cells with a solvent mixture comprising a halogenated hydrocarbon and an alcohol, (ii) separating the insoluble residue from the liquid extract, (iii) evaporating the liquid extract to obtain a solid residue, (iv) extracting the solid residue with chloroform to obtain a chloroform solution of said immunostimulating agent and (v) separating this chloroform solution from any undissolved solid residue and (b) a pharmaceutically acceptable carrier. The splenic cells may be mice splenic cells which optionally have been infected with Listeria monocytogenes bacteria. The bacterial cells used to prepare the extract may be cells of Listeria monocytogenes, Salmonella typhimurium, Escheria coli, Corynebacterium parvum, Mycobacterium tuberculosis, Vibrio cholera, Bacillus subtilis. The extraction solvent mixture may be chloroform and methanol. The pharmaceutically acceptable carrier may be an injectable liquid carrier, e.g. saline. A saline dispersion is prepared by dispersing the chloroform solution in saline, bubbling a gas through the dispersion to evaporate the chloroform and homogenising the resulting dispersion. As an alternative to the injectable solution, the immunostimulating agent can be incorporated in a calcium phosphate gel.
