Abstract

1411386 Immobilized enzymes PFIZER Inc 18 Sept 1972 [12 July 1972] 43226/72 Heading C3H [Also in Division C2] Immobilized enzymes are obtained by reacting a hydroxy containing ethylenically unsaturated monomer with a cyanogen halide preferably bromide in aqueous solution at an alkaline pH, reacting the neutralized reaction mixture containing the resulting monomercyanogen halide product with an aqueous solution of the enzyme and polymerizing the monomer-enzyme adduct in the presence of a cross-linking agent alone or in the presence of another ethylenically unsaturated monomer and catalyst, said hydroxy containing monomer having the general formula wherein X is O or NR<SP>1</SP> and R<SP>1</SP> is hydrogen or alkyl containing from 1-6 carbon atoms, A is alkylene containing from 2-6 carbon atoms or (YO) n Y where Y is an ethylene or propylene residue and n = 1 or 2, R 1 is hydrogen or in which X and A are as above defined, R 2 is hydrogen, methyl, chlorine or in which X and A are as above defined, and where when R 1 is hydrogen, R 2 may be hydrogen, methyl, chlorine or and when R 1 is R 2 may be hydrogen or methyl. Preferred comonomers are, when R 1 = hydrogen, acrylamide, N,N-dialkyl acrylamide, lower alkyl esters of acrylic, methacrylic or α-chloroacrylic acid, acrylonitrile, methacrylonitrile or styrene, and when R 2 = hydrogen or methyl, are vinyl chloride, vinylidene chloride, vinyl acetate, allyl proprionate, methallyl butyrate, methyl vinyl ether, N-vinylpyrrolidone, styrene or vinyl pyridine. Specified enzymes include penicillin acylase, urease, amyloglucosidase, fumarase or subtilopeptidase A from Bacillus subtilis ATCC 21839. The penicillin acylase may be derived from Escherichia coli ATCC 9637 or Aspergillus flavus Link ATCC 13608.
