Abstract
1400714 Mixing apparatus G D SEARLE & CO Ltd 13 Sept 1972 [28 Sept 1971] 45134/71 Heading B1C A device for making an isokinetic liquid density gradient in a centrifuge tube 27 comprises a closed mixing chamber 40 containing initially a liquid (sucrose) of lower concentration which is displaced by liquids of higher concentration into the tube 27 through a capillary tube 25, the open end of which is close to the bottom of the tube 27. The more concentrated liquid is supplied from a burette 44 having an optical level sensor 45 which closes a pinch-cock 43 and may stop any pump included in lines 26, 46, when sufficient liquid has been supplied to fill the tube 27 to the desired level; as the concentration in chamber 40 continuously increases, the more dense liquid will continuously displace the less dense upwards as it enters tube 27. Alternatively a sensor-associated with tube 25 may directly sense the level in tube 27. There is a magnetic stirrer 41 in chamber 40. The tube 27 is secured and centred between conical recesses in fixed and sliding blocks 28, 29 respectively and the capillary tube 25 is supported by a sliding block 23 which can be raised slowly to avoid turbulence by a variable speed motor (13, Figs. 1-3 not shown). A plurality of gradient-making devices may be mounted on a common stand having a common motor to raise all the capillary tubes simultaneously.
