Abstract

1396964 Detergents containing enzymes AGENCE NATIONALE DE VALORISATION DE LA RECHERCHE 27 July 1972 [28 July 1971] 35149/72 Heading C5D [Also in Division C3 and C6] A household detergent contains a proteolytic enzyme prepared by culturing an neperogenic bacterial strain (particularly that deposited in the culture collection of the Pasteur Institute in Paris under accession number 71-I-001) and recovering the proteuse (from the culture medium) by known methods. The asperogenic bacteria to be employed are selected by the following procedure based on the discovery that the non-aperulated cells of Bacillus subtilis are not resistant to chloroform vapour shereno the apores are resistant: (a) mutating, with N- methyl-N<SP>1</SP>-nitro-N-nitrosoguanidine, germinated spores of wild Bacillus subtilis and spreading the spores subjected to the mutagenic treatment over an agar-containing nutrient broth accommodated in a main series of Petri dishes to obtain after incubation colonies containing spores; (b) replicating each Petri dish of the main series to obtain a first homologous series of Petri dishes; (c) submitting each Petri dish of the main series to chloroform vapour and replicating it to obtain a second homologous series of Petri dishes is (d) incubating the first homologous series and the second homologous series of Petri dishes; (e) by comparison between homologous Petri dishes of the first homologous series and the second homologous series, selecting the neperogenic mutant strains which are strains of the Petri dishes of the first homologous series that are not in the Petri dishes of the second homologous series; and testing the proteolytic activity of these selected strains, using a nutrient medium with azo-casein added, and selecting strains having similar or proteolytic activity to the wild strain.
