Abstract

1392671 Citrates of 2-(N-hexamethyleneimino)-ethyl-(3-thienyl)cyclohexylacetate INNOTHERA 22 March 1973 [2 March 1973] 13910/73 Heading C2C The invention comprises the citrates of the ester of general Formula I which are prepared by reacting the amino ester of Formula I with citric acid. The compounds may be racemates or optical isomers depending on the optical nature of the starting amino ester. The amino ester of Formula I may be prepared by (a) condensing (3-thienyl)-cyclohexylacetic acid with a hexamethylene-imino-ethyl halide, (b) condensing a Ag or alkali metal salt of (3-thienyl)-cyclohexylacetic acid with a hexamethylene-imino ethyl halide or (c) by transesterification between hexamethylene-imino ethanol and an alkyl (3-thienyl)-cyclohexyl-acetate. ( + )- and ( - ) - 2 - (N - hexamethyleneimino) - ethyl - (3 - thienyl) - cyclohexylacetate are prepared by fractional crystallization from the quinine salts of the racemic acid obtained by heating together the acid and base followed by hydrolysis of the salt to obtain the free acid. Pharmaceutical compositions having spasmolytic, vasodilatory, analgesic anti-ischaemic activity for oral, rectal, or parenteral application comprise a citrate of a compound of Formula I together with a pharmaceutical carrier.
