Abstract
1389858 3 - Fluoro - D - alanine derivatives MERCK & CO Inc 13 June 1972 [18 June 1971 3 Feb 1972] 27621/72 Heading C2C The invention comprises carboxyl-protected derivatives and amino-protected derivatives of 3-fluoro-D-alanine or deutero analogues thereof, and a process comprising treating such derivatives with a cleaving agent to form 3-fluoro-D- alanine or its deutero analogue. The term "carboxy-protected derivatives" means compounds in which the carboxy group has been converted to an amide, substituted amide, a nitrile or an ester, such as the methyl, ethyl, tbutyl, benzyl or p-nitrobenzyl ester. The term "amino-protected derivative" means a compound in which the amino group is monoacylated or diacylated, is converted to a urethane, is alkylated or aralkylated or is converted to an isocyanate group. The following derivatives of 3-fluoro-D-alanine are described: N - benzyloxycarbonyl-; N - 1 - menthoxyacetyl-; benzyl ester; N,N-dibenzyl-; N-carbobenzoxy- and its l-α-phenylethylamine salt; N-(D-α- methylbenzyl- and corresponding 2-deutero derivative; N-benzoyl- and its 2,3-dideutero derivative; and N-ethoxycarbonyl derivatives. Other derivatives and intermediates described are: N - (D - α - methylbenzyl) - # - fluoroacetaldimine; 2 - (D - α - methylbenzylamino)- 3 - fluoro - D - propionitrile; 2 - (D - α- methylbenzylimino) - 3 - fluoropropionic acid; methyl 2 - methoxycarbonyl - 3 - fluoropropionate; methyl 2-carboxy-3-fluoropropionate; and L-α- methylbenzylamino salts; methyl S-2-carboxy- 3 - fluoro - propionate; methyl S - 2 - isocyanato - 3 - fluoropropionate; methyl S - 2 - aminocarbonyl - 3 - fluoro - propionate; 2 - carboxy- 3 - fluoropropionitrile; R - 2 - carboxy - 3 - fluoropropionitrile; R-2-azidocarbonyl-3-fluoropropionitrile; S - 2 - isocyanato - 3 - fluoropropionitrile; D - 2 - amino - 3 - fluoro - propionamide.
