Abstract

1388312 Arabinofuranosyl adenine-3<SP>1</SP>,5<SP>1</SP>- cyclic monophosphate ICN PHARMACEUTICALS Inc 2 Nov 1972 50557/72 Heading C2P The invention comprises 9-beta-D-arabinofuranosyl adenine-3<SP>1</SP>,5<SP>1</SP>-cyclic monophosphate and pharmaceutically acceptable salts thereof which may be prepared by reacting 8-halo- 3<SP>1</SP>,5<SP>1</SP>-cyclic adenosine monophosphate in a basic solution with an appropriate leaving group acid chloride to replace the H in the 2<SP>1</SP>-OH group with a leaving group (LG), either (a) reacting the compound obtained with an acetic acid/sodium acetate solution to produce Z<SP>1</SP>-OLG - 8 - hydroxy - adenosine - 3<SP>1</SP>,5<SP>1</SP> - cyclophosphate and subjecting this to a base catalysed cyclization reaction to produce 8-2<SP>1</SP>-anhydro- 8 - oxy - 9 - beta - D - arabinofuranosyladenine - 3<SP>1</SP>,5<SP>1</SP> - cyclic monophosphate or (b) reacting the compound containing the 2<SP>1</SP>-OLG group with methanolic ammonia to produce the above 8-2<SP>1</SP>-anhydro compound, reacting this with liquid H 2 S to produce 8-mercapto-9-beta- D - arabinofuranosyladenine - 3<SP>1</SP>,5<SP>1</SP> - cyclic monophosphate and finally subjecting this to catalytic reductive desulphurization. The compound of the invention and its pharmaceutically acceptable salts may be used as the active ingredients in conventional antiviral compositions.
