Abstract
1385472 6-α-Bromoarylacetamido penicillin 1-oxides GLAXO LABORATORIES Ltd 15 Feb 1972 [24 Feb 1971 22 Nov 1971] 5375/71 and 54170/71 Heading C2C 6-α-Bromoarylacetamido penicillin 1-oxides, preferably compounds of general formula wherein R<SP>1</SP> is aryl and R<SP>2</SP> is hydrogen or a carboxyl group other than acyloxymethyl are prepared by brominating a 6-arylacetamido penicillin 1-oxide with a free radical brominating agent. Suitable free radical brominating agents include bromine, an N-bromoamide, an N-bromoimide and 1,3,5-tribromo-1,2,4-triazole and bromination may be initiated by azobisisobutyronitrile or a peroxide. Other means of initiating bromination include irradiation by ultraviolet, visible light or γ-rays. The bromopenicillins may be subjected to a ring enlargement reaction whereby a 3-methylcephem compound is formed. The bromine atom of the cephem or penicillin compounds may be replaced by other nucleophiles. Ampillin and cephalexin may be prepared from the bromo-penicillins and bromocephalosporins respectively.
