Abstract

1383765 α - Guanidino - benzylpenicillins BEECHAM GROUP Ltd 6 Sept 1972 [4 Aug 1971] 36667/71 Heading C2C Novel penicillins having the general Formula (II) and salts thereof, wherein R 1 is hydrogen o hydroxyl, R 2 is hydrogen or methyl, and R 3 is an acyloxyalkyl group having 3 to 8 carbon atoms are prepared by reacting an acid addition salt of a compound of Formula IV with a compound of Formula V wherein R 2 and R 3 are as defined above, R 4 is hydrogen or an optionally protected hydroxy group, Y is hydrogen or a silyl group and X is a group which is readily displaceable to form an amide link, and thereafter if necessary removing any silyl group or protecting group. Alternatively to penicillins (II) wherein R 3 is pivaloyloxymethyl may be prepared by esterifying the appropriate penicillanic acid with pivaloyloxymethanol or an esterifying derivative thereof. The compounds IV are prepared by reacting an appropriate hydroxyphenyl α-aminoacetic acid with O-methyl isourea hydrochloride. The compounds (IV) are then converted to their hydrochloride salts. The penicillins (II) or their pharmaceutically acceptable salts are used to prepare antibiotically active pharmaceutical compositions.
