Abstract
1368731 9,11-Epoxy-spirolactones and related steroids G D SEARLE & CO 18 Jan 1973 [24 Jan 1972] 2574/73 Heading C2U Novel steroids of the formulae wherein Z is CO or CHOH or CHO (C 1-7 alkyl) in either configuration and X is H, ammonium or one equivalent of an alkali or alkaline earth metal are prepared from 9α,11α-epoxy-4<SP>1</SP>,5<SP>1</SP>-dihydrospiro[androst - 4 - ene - 17,21 - (3<SP>1</SP>H)- furan]-3,5<SP>1</SP>-dione by (a) catalytic hydrogenation to give the 5#-compound (Ia) wherein Z is CO, and when desired conversion of this to the 3,3- dimethyl ketal and reduction and hydrolysis of this with lithium and ammonia to give the 5#- compound (Ia) wherein Z is CHOH which may subsequently be etherified using an alkanol in the presence of acid; or (b) reduction of the same starting material with lithium and ammonia to give a mixture of the 5α-compounds (Ia) wherein Z is CO and CHOH, of which the latter may subsequently be etherified. Products wherein Z is CO may be converted to alkali metal salts (Ib) by reaction with alkali metal hydroxides and these salts may be converted to the free acids and other salts by standard procedures. The novel steroids are diuretic and hypotensive agents. Solutions in corn oil and other inert solvents, e.g. water, are referred to.
