1359226 Medicament base compositions SYNTEX CORP 8 Sept 1971 [29 Oct 1970] 41932/71 Heading A5B A composition, useful as a medicament base, consists essentially of (a) from 0.1 to 30 weight percent propylene carbonate; b) from 30 to 99.5 weight percent petrolatum or polysiloxane or mixtures thereof; c) compatible cosolvent and coupling agent, the concentration of which in combination with the propylene carbonate is from 0.5 to 70 weight percent; (d) from 0 to 45 weight percent pharmaceutically acceptable surfactant; (e) from 0 to 20 weight percent thickener: and (f) from 0 to 25 weight percent penetrant. The pentrolatum may be a paraffinic mineral oil or paraffin wax; and the polysiloxane may have organic moieties such as C 1-8 alkyl, C 1-8 alkenyl, phenyl, alkyl-substituted phenyl, and phenyl alkyl (e.g. benzyl) attached to the polysiloxane, dimethylpolysolixane being preferred; the compatible cosolvent and coupling agent may be chosen from mono-, di- or poly-hydric alcohols, alkyl ethers of glycols, polyethylene or polypropylene glycol, esters of aliphatic mono- and di-basic acids and mono-, di- and polyhydric alcohols, sugar alcohols or a sterol such as cholesterol; the surfactant may be anionic, cationic, amphoteric or nonionic; the thickener may be colloidal alumina or silica, alginic acid or derivatives thereof, carboxyvinyl polymer, cellulose derivatives (such as methyl cellulose, hydroxyethyl cellulose or sodium carboxymethyl cellulose), gelatin, gums (such as agar, tragacanth, acacia or guar gum), egg yolk, lecithin, pectin, thixcin or resins (such as ethyleneoxide polymers); and the penetrant may be a dialkyl sulphoxide, dimethyl-formamide, dimethylacetamide, tetrahydrofuran or tetrahydrofurfuryl alcohol. The composition may also contain beeswax, spermaceti, paraffin waxes, fatty acids, alcohols and amides; a penetration aid such as squalene or acetylated lanolin fractions and/or a preservative. The composition may contain a therapeutic agent for topical application such as an antibiotic, e.g. oxytetracylcline, chlorotetracycline, streptomycin, bacitracin, chloramphenicol or tyrothricin; a steroid; an antihistamine such as prophenpyridamine maleate or diphenhydramine hydrochloride; an anaesthetic such as benzocaine or lidocaine; an antibacterial such as iodine, nitrofurazone, sulphanilamide and derivatives, or benzalkonium chloride; a fungicide such as undecylenic acid; coal tar, balsum Peru, ammoniated mercury, chrysarobin, ichthammol or sulphur.
