1356080 Antibiotic A-130-A SHIONOGI & CO Ltd 28 Dec 1972 59825/72 Heading C2A The novel antibiotic A-130-A, characterized as being an amorphous colourless powder melting at 87‹ to 92‹ C., containing the elements carbon (66À47%), hydrogen (9À15%) and oxygen (24À07%), having an optical rotation [α]<SP>23</SP> D = + 64À5 degrees (C = 1À0 in CHCl 3 ), a molecular weight of about 850, and an infra-red absorption spectrum and nuclear magnetic resonance spectrum as shown above in Figs. 1 and 3 respectively, is produced by aerobically cultivating Streptomyces hygroscopicus (ATCC 21840) or an antibiotic A-130-A-producing mutant thereof in an aqueous nutrient medium and isolating said antibiotic from the cultured broth. The antibiotic is an acid compound forming base-salts, e.g. with sodium or ammonium, having a proposed molecular formula C 47 H 78 O 13 , an ultra-violet absorption peak at 234À5 mÁ (# = 14,200), and having α,#-unsaturated ketone and polycyclic polyether structures in the molecule. It decolorizes KMnO 4 solution, gives a positive Dragendorff test and a negative FeCl 3 reaction, and is soluble in most organic solvents but insoluble in water. The sodium salt is a white crystalline substance melting at 227‹ to 231‹ C., having a proposed molecular formula C 47 H 77 O 13 Na. The antibiotic is isolated either from the whole culture broth or from the separated broth and mycelium, by extraction with an appropriate organic solvent. The whole broth is acidified, filtered after addition of a diatomaceous earth absorbent, and the filtrate is extracted with a water-immiscible solvent, e.g. ethyl acetate, while the filter cake is extracted with a water-miscible or -immiscible solvent, e.g. acetone or ethyl acetate. The extracts are combined, concentrated, and either treated with water to precipitate the crude antibiotic or subjected to purification on a silicic acid column, the product being finally recrystallized from ether to give the pure antibiotic. The antibiotic A-130-A has anti-bacterial and anthelmintic properties and may be used to treat or prevent avian coccidiosis in poultry, optionally after admixture with a poultry feed or pharmaceutical carrier.
