Abstract

1344727 Chondroitin sulphates and compositions thereof L M MORRISON 19 April 1971 [15 April 1970] 27157/71 Heading A5B [Also in Division C3] Chondroitin sulphates A and C capable of causing a prolongation of plasma thrombus formation time 6 to 12 hours after administration in rabbits according to a defined procedure, are used orally (tablets) or by injection for preventing or treating atherosclerosis, particularly in human beings. A unit dose may amount to 0.5-10 g of the so-called biologically active chondroitin sulphates A and C. The compounds may be prepared from bovine trachea or shark cartilage, respectively, by a method broadly comprising defatting the trachea or cartilage, drying and grinding the defatted material, solubilizing the defatted material in a buffered medium containing a proteolytic enzyme and enzyme activators, this mixture being maintained at a temperature of 62‹ Œ 3‹C for not less than 24 hours, precipitating the supernatant with a fat solvent, dissolving the precipitate in isotonic saline to form a weak solution, adding to the solution small amounts of an oxidising agent until there is no further reaction, allowing the solution to stand or adding a small volume of formaldehyde or methanol, followed by a fat solvent, recovering the chondroitin sulphate A or C, testing it for biological activity/prolongation of plasma thrombus-formation time) and bringing it into a form suitable for administration.
