Abstract

1323902 Flocculating DREW CHEMICAL CORP 21 July 1970 [29 July 1969] 35243/70 Heading C1C [Also in Divisions C2 and C3] A process for removing suspended contaminants from a fermentation broth containing a desired product (e.g., protease, amylase, lipase, penicillin, tetracycline or citric acid) comprises flocculating the contaminants with (a) at least one polyamine prepared by reacting a compound of formula where Q = H, C 1 -C 4 alkyl, aminoalkyl with 1-4 C in the alkyl portion, x = 2-8, y = 1-4, with a compound of formula where L and M = Cl, Br, I or and/or (b) at least one polyvinyl imidazoline which has a pendant cyclic structure of where T = [(CH 2 ) v CH 3 ] or H, R = H or C 2 -C 6 alkyl, v = 1-3, and separating (e.g., by filtering or centrifuging) the flocculated contaminants. The desired product may be recovered from the broth by precipitation, solvent extraction, ion exchange adsorption or volatilization. A filter aid (e.g., diatomaceous earth) may be used in the process. Polymer (b) may be prepared by reacting a polymer prepared from an unsaturated nitrile (e.g., acrylonitrile, methacrylonitrile or cyanostyrene) alone or in combination with an ethylenically unsaturated monomer (e.g., butadiene, 2-pentene, vinyl chloride or acrylamide), and an amino compound (e.g., ethylene, propylene, hexamethylene or N-butylethylene diamine). In the Examples a protease broth is treated with a polymer prepared from polyhexamethylene polyamine with epichlorohydrin (Example IX); a tetracycline broth with a polymer prepared from polyhexamethylene diamine and epichlorohydrin (Example X); and an amylase broth with a homopolymer of acrylonitrile reacted with 1,2 diaminohexane (Example XI). Example VII discloses a polymer (b) prepared from polyacrylonitrile (M.W. 10,000) and 1,2 diaminohexane and Example VIII, a polymer (b) from a copolymer of cyanostyrene and butadiene, and ethylene diamine.
