1322339 2 - (2 - Imidazolin - 2 - yl) - benzyl alcohols AMERICAN HOME PRODUCTS CORP 2 Oct 1970 [10 March 1970] 46941/70 Heading C2C Novel 2 - (2 - imidazolin - 2 - yl) - benzyl alcohols of the general formula wherein R<SP>1</SP> is a phenyl, mono- or di-halophenyl, mono- or di-C 1-4 alkyl (not t-butyl) phenyl, trifluoromethylphenyl, mono- or di-C 1-6 alkoxyphenyl, thienyl, pyridyl, furyl or 5,6,7,8-tetrahydro-2-naphthyl group, R<SP>2</SP> is a hydrogen or halogen atom or an amino, C 1-4 alkyl (not tbutyl) amino, C 1-4 alkyl (not t-butyl) or C 1-6 alkoxy group, R<SP>3</SP> is a hydrogen atom when R<SP>2</SP> and R<SP>3</SP> are dissimilar and when R<SP>2</SP> and R<SP>3</SP> are the same they are both selected from hydrogen and halogen atoms and C 1-4 alkyl (not t-butyl) and C 1-6 alkoxy groups, and R<SP>4</SP> and R<SP>5</SP>, which are attached to the same carbon atom, are each a C 1-4 alkyl (not t-butyl) group, and acid addition salts thereof are prepared by reduction of a 2,3 - dihydro - 5H - imidazo[2,1-a]isoindol- 5 - ol or its tautomer of the respective general formulae followed optionally by salification of the product. 2,3 - Dihydro - 5H - imidazo[2,1-a]isoindol - 5 - ols of the second general formula above are prepared by reaction of a benzoic acid and an ethylenediamine of the respective general formulµ reaction of the resulting 1,2,3,9b-tetrahydro-5H- imidazo[2,1-a]isoindol-5-one of the general formula with a sulphonyl chloride of the general formula R<SP>7</SP>SO 2 Cl, wherein R<SP>7</SP> is an optionally substituted hydrocarbon group, and treatment of the resulting sulphonyl derivative of the general formula with a strong acid, followed by a base. Pharmaceutical compositions having antiinflammatory sctivity comprise, as active ingredient, a 2-(2-imidazolin-2-yl)-benzhydrol of the first general formula above or an acid addition salt thereof, in association with a pharmaceutically acceptable carrier, and may be administered orally or parenterally.
